花生红衣水提物对D-半乳糖衰老小鼠学习记忆能力的影响

【摘要】 目的观察花生红衣水提物对D-半乳糖衰老模型小鼠学习记忆能力的影响。方法 60只雄性昆明小鼠被随机分为空白对照组、衰老模型组、吡拉西坦阳性对照组、花生红衣水提物高、中、低(650.0、325.0、162.5 mg/kg)剂量组。空白对照组颈背部皮下注射0.5 mL生理盐水,其余5组均注射等体积2.5%的D-半乳糖,qd,连续42 d复制小鼠亚急性衰老模型。给药各组在造模同时分别灌胃给予相应药物,42d后通过跳台实验和水迷宫实验检测小鼠学习记忆能力;行为学检测结束后处死小鼠,计算脑、脾脏指数;ELISA法检测小鼠脑组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果与空白对照组小鼠比较,模型组小鼠体质量增加量明显降低,脑、脾指数显著降低;在跳台实验中潜伏期明显缩短,错误次数增多;在水迷宫实验中潜伏期明显延长,错误次数明显增多;脑组织内SOD活性下降,MDA含量增加。与模型组相比,吡拉西坦组和花生红衣各剂量组小鼠体质量增加量升高,脑、脾指数显著性增加,学习记忆能力提高,还可不同程度提高脑组织SOD活性,降低MDA含量。结论花生红衣水提物可改善D-半乳糖致衰老小鼠的学习记忆能力,作用机制可能与提高机体抗氧化能力和免疫活性有关。

【Abstract】 Objective To study the effect of aqueous extract from peanut red skin on learning and memory function in senile mice induced by D-galactose.Methods60 Male kunming mice were randomly divided into normal control group,aging model group,piracetam positive control group(2 mg/kg),and high,middle and low dose of peanut red skin aqueous extract groups(650.0 mg/kg,325.0 mg/kg,162.5 mg/kg). Mice in normal control group were subcutaneously injected with0.5 mL saline in cervical back,while those in other groups were subcutaneously injected with the same volume of 2.5% Dgalactose once daily for 42 days. The medicine was given at the same time. After 6 weeks,the learning and memory ability of mice were tested with step-down test and water maze test. The mice were killed after the behavior test,and the indexes of brain and spleen were calculated. ELISA method was used to detect the activity of superoxide dismutase(SOD)and the content of malonaldehyde(MDA)in brain tissue.ResultsCompared with normal control group,for aging model mice,the gain weights were decelerated relatively,the indexes of brain and spleen were decreased significantly;the number of errors was significantly increased,and the latency was obviously shortened in step-down test;the incubation period and the number of errors were significantly increased in the water maze test;the activity of SOD was reduced and the contents of MDA was raised. Compared with model group,mice in high,middle and low doses of peanut red skin water extract groups and piracetam positive control group showed increased weight gains,elevated the brain and spleen indexes,enhanced the learning and memory ability,improved the activities of SOD and decreased the contents of MDA.ConclusionThe peanut red skin water extract can improve the learning and memory ability of D-galactose-induced aging mice,and its mechanism may be related to the improvement of immunity and antioxidant capacity.
