药根碱促进N2aAPP细胞线粒体自噬的初步证据

【摘要】 目的探讨药根碱作用于AD细胞模型后,其线粒体自噬的超微结构变化。方法以N2a细胞作为对照组,N2aAPP细胞作为实验组,采用不同浓度的(5μmol/L和10μmol/L)药根碱分别作用于N2aAPP细胞和N2a细胞,24 h后收集细胞,进行活性氧检测和透射电镜观察。结果 N2aAPP组活性氧量最高,多于N2a组,差异有统计学意义(P<0.01);加药根碱处理24 h后,N2aAPP+5μmol/L JAT组和N2aAPP+10μmol/L JAT组活性氧均有减少,与N2aAPP组比较,差异有统计学意义(P<0.01)。与对照组N2a细胞组相比,N2aAPP细胞线粒体自噬的溶酶体与自噬小体较少,与N2aAPP细胞组相比,N2aAPP+5μmol/L JAT组和N2aAPP+10μmol/L JAT组的自噬溶酶体和自噬小体数量则相对增多。结论药根碱可能促进N2aAPP细胞的自噬溶酶体和自噬小体的增加,以促进线粒体自噬的降解。

【Abstract】 Objective To investigate the ultrastructural changes of mitochondrial autophagy in AD cell model in-duced by Jatrorrhizine(JAT).MethodsN2 a cells were taken as the control group and N2 aAPP cells as the experimental group. JAT were applied to N2 aAPP cells and N2 a cells at different concentrations(5 μmol/L collected 24 h later and observed by transmission electron microscopy.ResultsCompared with the N2 a cellsgroup,the num-ber of mitochondrial autophagolysosome and autophagosomes in the N2 aAPP group was lower(P<0.01). And the number of autophagolysosome and autophagosomes in the N2 aAPP+5 μmol/L JAT group and the N2 aAPP+10 μmol/L JAT group was higher than that in the N2 aAPP group.ConclusionJAT may promote the increase of autophagosome and autophagosome in N2 aAPP cells to promote the degradation of mitophagy.
