大鼠急性心肌梗死后血浆miRNA-1变化的研究
The change of plasma miRNA-1 in acute myocardial infarction rat model
【摘要】 目的研究利用miRNA-1诊断急性心肌梗死,在分子生物学层面拓展急性心肌梗死法医病理学诊断指标方法通过大鼠AMI动物模型,利用实时定量PCR技术检测急性心梗发生后血浆miRNA-1表达变化水平,并与免疫组化指标Connexin 43进行对比;利用实时定量PCR技术检测死亡早期miRNA-1变化趋势结果 AMI发生30min,血浆miRNA-1含量开始升高,AMI发生1h、2h后,血浆中miRNA-1相对含量平均值分别是对照组的3倍、5倍;而免疫组化指标Connexin 43在AMI发生30min后的表达部位发生改变,表达量升高;在死后稳定性方面,大鼠死亡0h、3h、6h、12h后,血浆miRNA-1含量没有统计学差异(p> 0.05),但在24h后,血浆miRNA-1明显升高,其平均值约为0h组的42倍(p <0.05)。结论在心梗发生早期,血浆miRNA-1含量特异性升高,并能在死亡早期保持稳定,有望作为法医病理诊断急性心肌梗死辅助分子指标。
【Abstract】 Objective to study the potentiality of utilizing mi RNA-1 to diagnose acute myocardial infarction, expanding the possible medicolegal biomarkers of AMI Method to detect the level of plasma mi RNA-1 of AMI rats by q RT-PCR, compared to immunobiological marker Connexin 43, meanwhile to verify the change of plasma miRNA-1 during early postmortem interval using q RT-PCR Results plasma miRNA-1 started to increase when AMI happened 30 min, and the levels of plasma miRNA-1 significantly increased in the 1 h and 2 h infarction groups(3 and 5 folds respectively). Meanwhile, the expression and location alterations of Connexin 43 in immunohistological image were observed. Within 12 h of early postmortem, the plasma miRNA-1 showed no obvious changes. however, when comparing to 0 h postmortem sample, the level of plasma miRNA-1 increased significantly 42 folds after 24 h postmortem sample. Conclusion we observed plasma miRNA-1 to be highly specific to acute myocardial infarction as well as be stable to early postmortem intervals, which could be a potential biomarker for forensic diagnosing AMI.
