全自动磁微粒化学发光法快速筛查呋喃类药物残留

【摘要】 文章通过开发硝基呋喃类药物磁微粒化学发光试剂盒,建立了以磁微粒作为固相载体、化学发光作为检测信号的硝基呋喃类药物残留快速检测方法。结合全自动化学发光仪,用磁微粒化学发光酶联免疫法(MCLIA)同时对4种硝基呋喃类药物(呋喃西林代谢物(SEM)、呋喃唑酮代谢物(AOZ)、呋喃它酮代谢物(AMOZ)和呋喃妥因代谢物(AHD))在不同来源的鸡肉样、鱼样中的残留量进行定量检测,并与基于超高效液相色谱—三重四极杆串联质谱联用仪的国家标准方法的检测结果进行了对比。共检测鱼肉、猪肉、鸡肉和贝类样品等共计78个样本,与国家标准方法检测结果的符合率在98%以上。结果表明全自动微粒化学发光酶联免疫法具备有自动化程度高,多项目同时检测,检测效率高,检测结果准确可靠等优势,可用于硝基呋喃类药物残留的快速筛查。

【Abstract】 In order to develop a magnetic particle chemiluminescence reagent kit for nitrofurans,a rapid detection method was established for the nitrofurans residues,which was based on magnetic particles as solid phase carrier and chemiluminescence as detection signal.The samples from different sources of chicken and fish,were been simultaneously quantitative detected for four nitrofurans residues as furacillin metabolites(SEM),furazolidone metabolites(AOZ),furan-tazone metabolites(AMOZ) and furantoin metabolites(AHD),by magnetic particle chemiluminescence enzyme-linked immunoassay(MCLIA) combined with full-automatic chemiluminescence apparatus,and then compared the results with the national standard method,which based on ultra-high performance liquid chromatography-triple quadrupole tandem mass spectrometry.A total of 78 samples of fish,pork,chicken and shellfish were detected,and the coincidence rate,comparing the results with the national standard method’s,was over 98%.The full-automatic particle chemiluminescence ELISA had the advantages of high automation,multi-item simultaneous detection,high detection efficiency and accurate and reliable detection results,while it could be used for rapid screening of nitrofurans residues.
