酸凝乳的扫描电镜样品制备方法

【摘要】 本实验根据酸凝乳样品的特性,在常规扫描电镜制样方法的基础上,对样品的取材、固定和导电处理等几个重要环节进行了优化,比较了不同浓度的戊二醛固定液和不同喷镀条件的制样效果。结果表明:固定液戊二醛浓度为4%以及在真空度为9.3 Pa,溅射电流6~8 mA、喷镀时间4 min,间歇喷镀4次条件下,制备的酸凝乳样品的凝胶体中各有形物质保存完好,网状立体结构层次清晰。

【Abstract】 In this experiment, based on the characteristics of solidification yoghurt samples and the conventional SEM sample preparation method, several important links of sample sampling, fixation and conductive treatment were optimized. The effects of different concentrations of glutaraldehyde fixing solution and different spraying conditions on sample preparation were compared. The experimental results showed that the sample was fixed with glutaraldehyde solution with a concentration of 4%, and the conductive layer was sprayed as follows: vacuum degree 9.3 Pa, sputtering current 6-8 mA, spraying time 4 min, intermittent spraying 4 times. The solid substances in the gel of the solidification yoghurt sample were well preserved and the reticular three-dimensional structure was clear.
