玉屏风复合多糖的质量控制研究
Study on Quality Control of Yupingfeng Polysaccharides
【摘要】 旨在建立一种玉屏风复合多糖(Yupingfeng polysaccharides,YPF-P)的质量控制方法。提取纯化YPF-P,采用高效凝胶色谱(HPLC-GPC)法测定YPF-P的分子质量。流动相:超纯水,流速:0.7 mL/min,柱温:35℃;薄层色谱(TLC)法测定多糖的单糖组成,使用硅胶G板二次展开,105℃显色45 min;MTT法对多糖作用后的CEF体外增殖能力测定,优选出YPF-P溶液中的最佳浓度,然后与其3种组方中药多糖进行比较试验。结果显示,YPF-P中Mw约为5 ku小分子多糖占其85%;YPF-P由果糖、D-葡萄糖、D-甘露糖、阿拉伯糖、D-半乳糖与、D-核糖、D-半乳糖醛酸组成;YPF-P的15.63μg/mL~1 000μg/mL各浓度的试验组均有显著促进CEF增殖的作用,其中1 000μg/mL浓度组促进增殖的效果最明显。4种多糖对CEF的增殖促进作用大小顺序为玉屏风复合多糖>黄芪多糖>白术多糖>防风多糖。研究结果表明,该方法稳定可靠,可以作为一种YPF-P的质量控制方法。
【Abstract】 This study aimed to establish a quality control method for Yupingfeng polysaccharides(YPF-P).YPF-P was Extracted and purified,and the molecular weight of YPF-P was measure by HPLC-GPC,mobile phase:ultrapure water,flow velocity:0.7 mL/min,column temperature:35℃.TLC was used to determine the monosaccharide composition of YPF-P,using two kinds of expansion agents to expand two times on the silica gel G plate,heating and developing the sample(105℃) for 45 min.MTT assay was used to test CEF proliferation in vitro.The best YPF-P concentration was selected,and then 4 kinds of polysaccharides were compared with the best concentration.The results showed:85% of polysaccharides from YPF-P was Mw≈5 ku small molecule polysaccharides; YPF-P consists of fructose,D-glucose,D-mannose,arabinose,D-galactose,D-ribose and D-galacturonic acid.The tested groups of 15.63 μg/mL-1 000 μg/mL of YPF-P could significantly promote the proliferation of CEF.The best concentration was 1 000 μg/mL.The effect size order of 4 polysaccharides on CEF proliferation was YPF-P>Astragalus polysaccharides>Atractylodes polysaccharides>Sposknikovan.The results showed that the method is stable and reliable,and can be used as a quality control method for YPF-P.
