重组酿酒酵母表达白斑综合征病毒VP28的保护性初探
Preliminary Study on Protective Efficacy of Recombinant Saccharomyces cerevisiae Expressing VP28 of White Spot Syndrome Virus
【摘要】 目的:构建重组酿酒酵母表达白斑综合征病毒VP28蛋白,并对其口服给药后的保护效率进行分析。方法:以白斑综合征病毒的vp28基因(基因库编号:FJ756456.1)作为研究对象,构建重组酿酒酵母S.cerevisiae EBY100/pYD1-VP28,通过SDS-PAGE、Western blot以及免疫荧光分析对重组酿酒酵母S.cerevisiae EBY100/pYD1-VP28的表达进行检测。以白肢虾(Penaeus vannamei)作为动物模型,通过白斑综合征病毒攻击实验,检测重组酿酒酵母S.cerevisiae EBY100/pYD1-VP28的保护效率。结果:口服重组酿酒酵母EBY100/pYD1-VP28后的对虾存活率为60%。结论:重组酿酒酵母S.cerevisiae EBY100/pYD1-VP28可以应用于对虾养殖业预防白斑综合征病毒的感染,潜藏着巨大的市场应用前景。
【Abstract】 Objective:To construct recombinant Saccharomyces cerevisiae(S. cerevisiae) expressing VP28 antigen protein of white spot syndrome virus(WSSV) and investigate its protective efficacy after oral administration. Methods:Recombinant S. cerevisiae EBY100/pYD1-VP28 was constructed based on vp28 gene of WSSV(gene accession No.FJ756456.1)as research subject and analyzed by SDS-PAGE, Western blot and immunofluorescence assay.Further, Penaeus vannamei was used as an animal model, protective efficacy of recombinant S. cerevisiae EBY100/pYD1-VP28 was determined by WSSV challenge. Results:Oral administration of recombinant S. cerevisiae EBY100/pYD1-VP28 could provide 60% of survival rate in Penaeus vannamei. Conclusion:Recombinant S. cerevisiae EBY100/pYD1-VP28 can prevent WSSV infection in shrimp culture industry and has a great potential for market.
