基于明胶-海藻酸钠复合水凝胶的挤压式3D细胞打印研究
Study of Extrusion Three-dimensional Cell Printing Based on Gelatin-Alginate Hydrogels
【摘要】 制备了明胶-海藻酸钠复合水凝胶,研究了不同配比下复合水凝胶的流变特性,并将其用于挤压式3D细胞打印。实验结果表明:在明胶、海藻酸钠的质量体积比分别为50、60 g/L时,复合水凝胶材料的打印性能较好,能避免喷头堵塞或材料从喷嘴滴落等问题;在气压为0.1~0.3 MPa、打印速度为10~20 mm/s、喷头直径为210~920μm条件下,可将打印出的细丝直径控制在300~1500μm,能按需打印出水凝胶薄膜或三维结构,且用复合水凝胶打印出的细胞在静态培养14天后仍存活90%以上。
【Abstract】 The gelatin-alginate hydrogels were prepared and its rheological properties were studied in different concentrations of gelatin and alginate for extrusion-bioprinting. The results indicated that when the mass-to-volume ratio of gelatin and alginate are 50 g/L and 60 g/L,the hydrogels gets a good printability,and the problems such as nozzle clogging or the tip droplet formation and dripping could obviously avoided. When the atmospheric pressure was 0.1 ~0.3 MPa,the printing speed was 10 ~20 mm/s and the nozzle diameter was 210~920 μm,the diameters of cylindrical filaments could be controlled in 300 ~1500 μm,and can print hydrogel film or three-dimensional structure on demand.The cells composite experiments showed that the cells kept their viability over 90% after 14 days.
