某市郊区机动车尾气PM2.5对小鼠骨髓细胞的微核试验研究

【摘要】 目的探讨某市郊区机动车尾气PM2.5对小鼠骨髓细胞的致突变性。方法采集某市郊区机动车尾气PM2.5并制备成不同浓度的颗粒物悬液。将受试健康昆明种小鼠50只随机分5组(n=10),阴性对照组(生理盐水)、阳性对照组(第13、15天,环磷酰胺100 mg/kg腹腔注射)、PM2.5低剂量组(5 mg/kg)、PM2.5中剂量组(10 mg/kg)、PM2.5高剂量组(15 mg/kg),采用暴露式气管滴注方式进行染毒,连续15 d。测定小鼠骨髓嗜多染红细胞(PCE)中的微核率的变化。结果各实验组小鼠骨髓细胞微核率比较差异有统计学意义(P<0.001),且染毒浓度与小鼠骨髓细胞微核率存在一定的剂量反应关系。结论 PM2.5使小鼠骨髓细胞微核率增加,有遗传毒性作用。

【Abstract】 Objective To investigate the mutagenicity of PM2.5 from motor vehicle exhaust in a suburb of a city on mouse bone marrow cells. Methods PM2.5 from vehicle exhaust was collected and prepared into particulate suspension of different concentrations. 50 healthy Kunming mice were randomly divided into 5 groups,including negative control group(normal saline),positive control group(13 th-15 th day,cyclophosphamide 100 mg/kg,intraperitoneal injection),low-dose PM2.5 group(5 mg/kg),medium-high-dose PM2.5 group(10 mg/kg),high-dose PM2.5 group(15 mg/kg).Exposed trachea infusion was used for poisoning for 15 consecutive days.The micronucleus frequency of polychromatic erythrocytes(PCE) in mouse bone marrow was measured.Results The difference of the micronucleus frequency in bone marrow cells had statistical significance(P<0.001),and the micronucleus frequency revealed dose effect relationship to a certain extent. Conclusion PM2.5 increased the micronucleus frequency of bone marrow cells in mice and had genotoxic effects.
