蚓激酶对TGF-β1诱导HK-2细胞Col Ⅰ、PAI-1、NOX4表达的影响

【摘要】 目的观察蚓激酶对转化生长因子-β1(TGF-β1)诱导人肾小管上皮细胞(HK-2)ColⅠ、PAI-1、NOX4表达的影响,以初步探讨其延缓肾纤维化的作用机制。方法 MTT法检测不同浓度的蚓激酶作用于HK-2细胞对细胞活性的影响;实验分为正常组、模型组、贝那普利组、蚓激酶低、中、高剂量组。分别采用Western blotting和荧光定量PCR检测ColⅠ、PAI-1、NOX4表达的情况。结果不同浓度的蚓激酶对HK-2细胞活性无明显影响;与模型组相比,蚓激酶各浓度治疗后均能明显下调ColⅠ、PAI-1、NOX4的表达(P <0.05)。结论蚓激酶可以调节TGF-β1刺激的HK-2细胞ColⅠ、PAI-1、NOX4表达,从而阻抑肾纤维化的发展。

【Abstract】 Objective To observe the regulation of lumbrukinase on the expression of type Ⅰ collagen(Col Ⅰ),plasminogen activator inhibitor-1(PAI-1), NADPH oxidase 4(NOX4) induced by transforming growth factor-β1(TGF-β1) in human renal tubular epithelial cells(HK-2), so as to further explore the ef fective mechanism of anti-renalfibrosis of the lumbrukinase. Methods MTT assay was used to determine the activity of HK-2 treated with different concentrations of lumbrokinase. The experiment was divided into normal group, model group, benazepril group, and lumbrukinase low, medium, high dose group. Western blotting and real-time PCR were used to detect the expression of Col Ⅰ, PAI-1 and NOX4. Results Dif ferent concentrations of lumbrokinase had no significant effect on the activity of HK-2 cells. Compared with the model group, the expressions of Col Ⅰ, PAI-1 and NOX4 were significantly reduced after treatment with different concentrations of lumbrokinase(P < 0.05). Conclusion Lumbrokinase may prevent the development of renal fibrosis by regulating the expression of Col Ⅰ, PAI-1 and NOX4.
