云斑白条天牛成虫不同组织部位DNA提取方法比较
Extraction Methods Comparison of Genomic DNA Extracted from Different Body Parts of Batocera lineolata Adult
【摘要】 分别采用试剂盒法、SDS-K法和CTAB法,提取了云斑白条天牛成虫的胸部、足部、触角和腹部四个组织部位的DNA。通过计算OD260/OD280和琼脂糖凝胶电泳检测法,对不同提取方法和不同组织部位提取的DNA质量进行了检测比较。结果表明,试剂盒法提取云斑白条天牛的胸部、足部、触角和腹部四个组织部位的DNA条带清晰,OD260/OD280均在1.80~1.86,DNA质量最高;SDS-K法提取DNA质量次之;CTAB法较差,基本无条带。不同组织部位提取的DNA,胸部质量最高,足部次之,其浓度由高到低顺序依次为,胸部>足部>触角>腹部。因此,提取云斑白条天牛成虫DNA时,建议试剂盒法从胸部提取,但样品量较少且稀缺时,从足部提取。
【Abstract】 Batocera lineolata adult genomic DNA is extracted from four body parts,which are thoraxes,legs,antennas and abdomens,by the methods of Kit,SDS-K and CTAB.The quality of DNA extracted from different body parts with different methods is tested by calculating the ratio OD260/OD280 and agarose gel electrophoresis method.The results show that DNA extracted from the thorax,leg,antenna and abdomen of Batocera lineolata by the kit method displays clear bands,with the ratios OD260/OD280 of1.80~1.86.Moreover,the quality of DNA isolated by kit method is the best,followed with that of DNA obtained by SDS-K,and the quality of DNA extracted by CTAB is bad without bands.For the quality of DNA extracted from different body parts,the thorax is the best,followed by leg.The sequence of DNA concentration extracted from different body parts is followed from high to low,thorax>leg>antenna>abdomen.It is suggested Batocera lineolata adult DNA extracted from thorax by the kit method,however,it is a good choice to extract DNA from legs of the rare samples.
