白脉软膏中重金属透皮吸收及体内蓄积情况研究

【摘要】 目的:以外用制剂白脉软膏为研究对象,探讨其所含重金属的透皮吸收特性及体内蓄积情况。方法:以Franz扩散池为透皮吸收实验装置,采用ICP-MS测定铅(Pb)、镉(Cd)、汞(Hg)、砷(As)、铜(Cu)、铬(Cr)、铁(Fe)、钡(Ba)、铝(Al)9种重金属在体外释放透皮吸收行为,以大鼠血液中重金属元素含量为监测指标,观察重金属在体内蓄积情况。结果:Cr、Fe、As、Ba、Pb 5种元素24h内的最大皮肤透过量分别为0.0153、0.6499、0.0228、0.6858、0.0207mg/kg。大鼠涂抹白脉软膏15、30、60d及停药15d后检测大鼠血液中重金属含量,Pb、Fe、Cd、As、Cu 5种元素在大鼠体内均无明显蓄积,且无剂量依赖性。结论:白脉软膏中常见几种重金属元素在体内无明显蓄积,且无剂量依赖性。

【Abstract】 Objective: To study the transdermal absorption characteristics of heavy metals and internal accumulation in Baimai Ointment which was used for external application. Methods: The percutaneous absorption behavior of heavy metals such as lead(Pb), cadmium(Cd), mercury(Hg), arseni(As), copper(Cu), chromium(Cr), iron(Fe), barium(Ba) and aluminum(Al) in vitro were determined by ICP-MS using Franz diffusion cell as a transdermal absorption experimental device. Using the content of heavy metals in rat blood as monitoring index, the accumulation of heavy metals in vivo was observed. Results: The maximum skin penetration of Cr, Fe, As, Ba and Pb were 0.0153, 0.6499, 0.0228, 0.6858, 0.0207 mg/kg at 24 h. The contents of heavy metals in blood of rats were measured after applying Baimai Ointment for 15, 30, 60 days and 15 days after withdrawal. Pb, Fe, Cd, As and Cu had no obvious accumulation in the body and no dose dependence. Conclusion: Several common heavy metals in Baimai Ointment have no obvious accumulation in the body and no dose dependence.
