益气化瘀中药通过调节NF-κB的活性抑制MMP-2、MMP-9的表达延缓系膜增生性肾小球肾炎的纤维化进程

【摘要】 目的:通过益气化瘀中药黄芪、水蛭有效成分对脂多糖诱导增生的系膜细胞(MCs)的作用,研究益气化瘀中药对系膜增生性肾小球肾炎(MsPGN)治疗作用的可能机制。方法:用脂多糖刺激MCs增生,再加入黄芪多糖、黄芪甲苷、毛蕊异黄酮、水蛭素处理后,用RT-PCR法及免疫荧光法检测各组细胞中NF-κB、MMP-2、M M P-9的转录水平及蛋白水平的表达情况。结果:脂多糖刺激后,M C s明显增生,用药后,各给药组NF-κB、MMP-2、MMP-9 mRNA及蛋白的表达水平(除黄芪多糖组及毛蕊异黄酮组的NF-κB外)较模型组均下调(P<0.05)。结论:黄芪、水蛭有效组分作用于增生的MCs,可以延缓肾小球纤维化的进程,其机制可能与NF-κB被抑制后引起MMP-2、MMP-9的表达下降,导致细胞外基质沉积减少有关。
【Abstract】 Objective: To study the possible mechanism of Chinese herbs of benefiting qi and resolving stasis on mesangial proliferative glomerulonephritis by observing the function of the effective components of astragalus and leech on proliferation of mesangial cells induced by lipopolysaccharide(LPS). Methods: The mesangial cells were stimulated to proliferate by LPS, and then treated with astragalus polysaccharide, astragaloside, calycosin and hirudin. The transcription and protein expression level of NF-κB, MMP-2 and MMP-9 were detected by RT-PCR and immunofluorescence. Results: The proliferation of mesangial cells were significantly stimulated by LPS. The mRNA and protein expression levels of NF-κB, MMP-2 and MMP-9 were decreased in each groups(P<0.05), in addition to the mRNA and protein expression of NF-κB in astragalus polysaecharide,calycosin group. Conclusion: The active components of astragalus membranaceus and leech acted on the hyperplastic mesangial cells can delay the progression of glomerular fibrosis. The mechanism may be that the decrease of MMP-2 and MMP-9 expression after the inhibition of NF-κB leads to the reduction of extracellular matrix deposition in renal cells.
