【摘要】 目的:通过HPLC指纹图谱结合化学计量学评价不同产地川牛膝的药材质量,为川牛膝的产地质量差异和评价提供参考依据。方法:采用高效液相色谱-二极管陈列检测器(HPLC-DAD)多波长切换法测定30批次不同产地及年份来源的川牛膝,并进行相似度评价和主成分分析。结果:选取了18个色谱峰作为指纹图谱共有峰,相似度评价得出以对照图谱R为对照,金口河产川牛膝(S1~S14)相似度均低于0.56,其余产地产川牛膝(S15~S30)相似度均大于0.84,而S1~S14川牛膝与其生成的对照图谱R1相似度均大于0.977,S15~S30号川牛膝与其生成的对照图谱R2相似度均大于0.930。以主成分分析得出的6个主成分对不同产地川牛膝进行综合评分并排名,且与杯苋甾酮含量排名存有差异。结论:本实验建立的对照图谱R1和R2可用以区分金口河产川牛膝和其它产地产川牛膝,仅以杯苋甾酮含量高低难以代表川牛膝的整体质量,而主成分分析对整体质量评价提供科学依据。

【Abstract】 Objective: HPLC fingerprint and chemometrics were used to evaluate the quality of Cyathula offinalis Kuan in different habitats, which provide reference for the quality difference and evaluate. Methods: HPLC-DAD multi-wavelength switching method was used to determine 30 groups of Cyathula offinalis Kuan from different origins and years, and carry out similarity evaluation and principal component analysis. Results: A total of 18 chromatographic peaks were selected as the common peaks of fingerprints, using the contrast chromatogram R as control, the similarity of S1~S14 sample(Jinkouhe origin) were less than 0.56 and S15~S30 sample(other origin) were higher than 0.84. The similarity between S1~S14 and its contrast chromatogram R1 were higher than 0.977, and the similarity between S15~S30 and its contrast chromatogram R2 were higher than 0.930. The6 principal components, which obtained from the principal component analysis, were scoring and ranking for Cyathula offinalis Kuan from different habitats, and there was a difference with the ranking of cyasterone content. Conclusion: The established control chromatogram of R1 and R2 can be used to distinguish the origins of Cyathula offinalis Kuan between Jinkouhe and other origin. Only the cyasterone content is difficult to represent the overall quality of Cyathula offinalis Kuan, but principal component analysis provides a scientific basis for it.
