一例牛急性感染伪狂犬病病毒的分离鉴定

【摘要】 山东东营地区某牛场发生以流涎、眼睑肿大、表现为神经症状为主的疾病,通过采集病牛鼻腔试子,提取病毒基因组DNA,感作乳仓鼠肾(BHK-21)细胞,经细胞传代培养出现明显特征病变。以伪狂犬病病毒(PRV)高度保守的gB基因扩增引物进行PCR扩增,获得特异性的DNA片段,连接PMD18-T载体,测序鉴定与PRV SC株同源性高达99%以上。确诊为牛感染伪狂犬病病毒。

【Abstract】 A disease in the cattle farm in Dongying, Shandong province, which is mainly caused by salivation, swollen eyelids, and manifested as neurological symptoms. Virus DNA genome was extracted from nasal fluid and brain tissue of dead bovine, and infected to baby hamster syrian kidney(BHK-21) cell line. The cell culture showed obvious characteristic lesion. The aim DNA fragment was obtained by PCR amplification with the highly conserved gB gene amplification primer of pseudorabies virus(PRV), and was connected to PMD18-T vector. The homology of PRV SC strain was more than 99% by sequencing. Then the diagnosis of the disease was pseudorabies virus infected bovine.
