温阳利水方对心力衰竭大鼠心功能及炎症、氧化应激反应的影响

【摘要】 目的探讨温阳利水方对心力衰竭大鼠心功能及炎症、氧化应激反应的影响,揭示温阳利水方治疗心力衰竭的作用机制。方法采用腹主动脉缩窄法制备大鼠心力衰竭模型,将大鼠随机分为假手术组、模型组、曲美他嗪组、温阳利水方低剂量组和温阳利水方高剂量组,灌胃给药12周后,测定各组大鼠的左室舒张末期内径(LVEDD)、左室收缩末期内径(LVESD)、左室射血分数(LVEF);测定大鼠血浆脑钠肽(BNP)、可溶性ST2(sST2)、半乳糖凝集素-3(Galectin-3)、C反应蛋白(CRP)、白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的表达;测定大鼠心肌组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPX)、血红素氧合酶(HO-1)的表达。结果治疗12周后,与模型组比较,曲美他嗪组及温阳利水方两组的LVEDD、LVESD、BNP、sST2、Galectin-3均下降,LVEF值均升高;温阳利水方高剂量组与曲美他嗪组相比,LVEDD、LVESD、BNP、sST2、Galectin-3均下降,LVEF值均升高,差异均有统计学意义(P <0.05);与模型组比较,曲美他嗪组、温阳利水方两组的CRP、IL-6、TNF-α的表达均有所降低,且温阳利水方高剂量组低于曲美他嗪组,差异均有统计学意义(P <0.05);与模型组比较,曲美他嗪组、温阳利水方两组的SOD、GPX、HO-1的表达均有所升高,且温阳利水方高剂量组高于曲美他嗪组,差异均有统计学意义(P <0.05)。结论温阳利水方可以通过降低心力衰竭大鼠CRP、IL-6、TNF-α的表达水平,同时提高SOD、GPX、HO-1的表达水平,来降低LVEDD、LVESD、BNP、sST2、Galectin-3的指标,升高LVEF值,从而保护心肌,最终改善心力衰竭大鼠心功能。

【Abstract】 Objective To investigate the effects of Wenyang Lishui decoction(WLD)on heart function,inflammation,and oxidative stress in rats with heart failure,and to reveal the mechanism of WLD in the treatment of heart failure.Methods The rat model with heart failure was established by abdominal aortic coarctation.The rats were randomly divided into sham operation group,model group,trimetazidine group,WLD low dose group and WLD high dose group.After 12 weeks of intragastric administration,left ventricular end-diastolic diameter(LVEDD),left ventricular end-systolic diameter(LVESD),and left ventricular ejection fraction(LVEF)were measured in each group.The expressions of plasma brain natriuretic peptide(BNP),soluble ST2(sST2),galectin-3,C-reactive protein(CRP),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were determinated.The expressions of superoxide dismutase(SOD),glutathione peroxidase(GPX),and heme oxygenase(HO-1)in myocardial tissue were determinated.Results After 12 weeks of treatment,compared with model group,LVEDD,LVESD,BNP,sST2 and Galectin-3 were decreased while LVEF was increased in trimetazidine group and WLD group.Compared with trimetazidine group,LVEDD,LVESD,BNP,sST2 and Galectin-3 were decreased while LVEF was increased in trimetazidine group and WLD high dose group(P <0.05).Compared with the model group,the expressions of CRP,IL-6 and TNF-αwere decreased in trimetazidine group and WLD group,which were higher in WLD high dose group than that in trimetazidine group(P <0.05).Compared with the model group,the expressions of SOD,GPX and HO-1 were increased in trimetazidine group and WLD group,which were higher in WLD high dose group than that in trimetazidine group(P <0.05).Conclusion WLD can reduce LVEDD,LVESD,BNP,sST2 and Galectin-3,and increase LVEF value by reducing the expressions of CRP,IL-6 and TNF-αand increasing the expressions of SOD,GPX and HO-1 in rats with heart failure,thereby protecting the myocardium and ultimately improving heart function in rats with heart failure.
