鼠妇有效部位镇痛的药效学研究

【摘要】 目的:考察鼠妇有效部位在口服给药方式下的镇痛作用,开展药效学研究,并初步探索其活性成分。方法:以昆明小鼠为实验动物,首先开展鼠妇水提物的急性毒性研究;其次,采用醋酸扭体模型考察鼠妇不同极性提取物的镇痛作用,筛选有效部位。基于实验结果,依次选用福尔马林法、热板法研究该有效部位的作用环节及特点,随后采用角叉菜胶致足肿胀以及醋酸致毛细血管壁通透性升高模型考察其抗炎活性。此外,采用液质联用技术分析鼠妇的活性成分。结果:鼠妇水提物在2~5 g/kg的剂量范围内未见明显毒性反应,在0.6~0.8 g/kg剂量范围内能够显著减少小鼠在醋酸扭体实验中的扭体次数,在0.4~0.8 g/kg剂量范围内能够缩短福尔马林实验(第二阶段)中的舔足时间,但未能延长其在热板实验中的潜伏时间。在炎症模型中,鼠妇水提物(0.4~0.8 g/kg)能够减少小鼠在角叉菜胶实验中的足肿胀程度,而在0.6~0.8 g/kg剂量范围内能够显著降低毛细血管壁通透性。液质联用实验结果表明,在氨基葡萄糖对照品和鼠妇水提物样品的HPLC谱图中,在相同的保留时间处均有峰型一致的色谱峰,且它们在MRM模式下具有相同的质谱行为。结论:鼠妇水提物具有抑制炎症性疼痛的药理作用,氨基葡萄糖是其重要的活性成分。

【Abstract】 Objective: To evaluate the antinociceptive effect of Armadillidium vulgare Latreille,after its bioactive part was screened from the extracts obtained by different polar solvents. Methods: Firstly,acetic acid-induced writhing tests were performed to investigate analgesic effects of different extracts after the acute toxicity test of the aqueous extract of this medicine. Secondly,we assessed antinociceptve properties of them through formalin and hot plate tests,then studied anti-inflammatory effects by carrageenan and acetic acid-induced increase in capillary permeability tests. Finally,we analysed the bioactive compound of the extract by HPLC. Results: Oral administration of aqueous extract of those medicinal animals did not elicit any observable acute toxic effects at the doses of 2. 0 ~ 5. 0 g/kg,and reduced the number of acid-induced writhes at 0. 6 g/kg and 0. 8 g/kg. 0. 4 ~ 0. 8 g/kg extract inhibited paw-withdrawal during the second phase of the formalin test,but not the hot plate test. This aqueous extract demonstrated a strong positive effect in carrageenan test at doses of 0. 4 ~ 0. 8 g/kg,and in vascular permeability test at 0. 6 g/kg and 0. 8 g/kg. Glucosamine was detected and identified in it using HPLC. Conclusions: This study shows that the aqueous extract has the low toxicity profile,antinociceptive and anti-inflammatory effects,providing scientific evidence of the efficacy of its traditional use in pain treatment. Furthermore,our data suggest that glucosamine contributes,at least in part,to the analgesic activity of the extract.
