朝藿定A在大鼠不同组织中含量测定方法的建立及其组织分布研究

【摘要】 目的建立朝藿定A在大鼠不同组织生物样品中的LC-MS/MS含量检测方法及其在组织分布研究中的应用。方法采用Agilent Eclipse XDB-C18柱(150 mm×2.1 mm,5μm);流动相:乙腈-0.1%乙酸水(32∶68);流速:0.3 m L·min-1;柱温:40℃;质谱采用多反应离子监测(MRM)方式负离子检测,用于定量分析的离子对为m/z 897.6→675.5。结果朝藿定A在1500 ng·mL-1线性范围内具有良好的线性关系,日内、日间精密度及稳定性良好,能满足生物样品检测的方法学要求。大鼠单次灌胃给予淫羊藿提取物后,朝藿定A主要分布在肝、肺和雌性生殖器官中,且体内组织分布存在明显的性别差异。结论该分析方法专属、灵敏、准确,适用于不同组织生物样品中朝藿定A浓度的测定,可应用于朝藿定A的组织分布研究。

【Abstract】 Objective To establish an LC-MS/MS method for determination of epimedin A in biological samples andits application in rat tissue distribution study. Methods Chromatographic separation was performed on an AgilentEclipse XDB-C18 column(150 mm × 2.1 mm,5.0 μm) at 40 ?C. Mobile phase consisted of acetonitrile-0.1%formic acid in water(32∶68),and the flow rate was 0.3 m L·min-1. Quantification was performed by multiple reactionmonitoring(MRM) mode and the selected monitor ion were m/z 897.6→675.5. Results The analyte showed goodlinearity over the concentration of 1-500 ng·m L-1. RSDs of intra-day and inter-day assays can meet the requirementsfor quantitative analysis of biological samples. Tissue distribution studies showed that epimedin A could rapidly andwidely distribute and effectively target to liver,lung and female reproductive system after single oral administrationof Epimedii herba extract to rats,and there were significant gender differences in tissue distribution. Conclusion The established method displayed good specificity, sensitivity and efficiency for quantifying epimedin A in ratbiological samples and can be applied for tissue distribution studies.
