【摘要】 目的研究芪丹方对5/6肾切除所致慢性肾衰竭(CRF)大鼠肾脏的保护作用及机制。方法 50只SD大鼠随机分为5组:假手术组,模型组,卡托普利组(5 mg·kg-1),芪丹方高、低剂量组(12.34,6.17 g·kg-1),每组10只。采用5/6肾切除法制备CRF大鼠模型。考马斯亮蓝法检测24 h尿蛋白;检测血清中尿素氮(BUN)、肌酐(Scr)的含量;HE染色观察大鼠肾脏组织病理学改变;流式细胞小球微阵列术(CBA)检测血清炎症因子白细胞介素-1α(IL-1α)、白细胞介素-2(IL-2)的含量;免疫组化法检测肾脏组织CD68的表达。结果与假手术组比较,模型组大鼠24 h尿蛋白及血清BUN、Scr、IL-1α、IL-2含量显著升高(P<0.05,P<0.01),肾组织CD68阳性表达显著增加(P<0.01)。与模型组比较,各给药组大鼠的24 h尿蛋白,血清Scr、IL-1α、IL-2含量,及肾组织CD68阳性表达均显著降低(P<0.05,P<0.01),芪丹方高剂量组的血清BUN显著降低(P<0.05)。结论芪丹方能有效改善CRF大鼠肾功能,减轻肾脏病理改变,其作用机制可能与通过减少肾脏巨噬细胞的浸润,下调IL-1α、IL-2等细胞因子表达相关。更多还原

【Abstract】 Objective This study is aimed to study the effects and mechanisms of Qi Dan decoction on 5/6 renal removal induced kidney failure in rats. Methods Totally 50 SD rats were randomly assigned to Sham group,model group,captopril group(5 mg·kg^-1) and Qi Dan decoction in high and low dose groups(12.34 g·kg^-1,6.17 g·kg^-1), 10 rats in each group. Kidney failure rats were prepared by 5/6 nephrectomy, and rats were given pharmacotherapy by gavage once a day for 4 weeks. Urine protein levels were measured by Coomassie brilliant blue weekly. The levels of blood urea nitrogen（BUN）,serum creatinine（Scr） were detected. Pathological changes were visualized by hematoxylin and eosin（HE） staining. The levels of interleukin-1α（IL-1α） and interleukin-2（IL-2） in serum were detected by Cytometric Bead Array. Evaluation of tubulointerstitial macrophage infiltration was performed by immunohistochemistry. Results 5/6 nephrectomy induced kidney failure rats had a decreased average value of weight and increased levels of urine protein,BUN,Scr,IL-1α and IL-2 which could be reversed by Qi Dan decoction. Similarly, Qi Dan decoction ameliorated pathological changes and tubulointerstitial macrophage infiltration induced in kidney failure rats prepared by 5/6 nephrectomy. Conclusion These results demonstrate that Qi Dan decoction has a good therapeutic effect on 5/6 nephrectomy induced chronic kidney failure rats and the effective mechanism is closely related to regulating inflammatory response.更多还原
