白藜芦醇通过TRPC1抑制血管平滑肌细胞增殖作用研究

【摘要】 目的研究白藜芦醇(Resveratrol,RSV)通过瞬时受体电位C1通道(transient receptor potential canonical-channel 1,TRPC1)来抑制血管平滑肌细胞(Vascular smooth muscle cells,VSMCs)增殖的机制。方法采用血管环培养法观察RSV对胎牛血清(fetal calf serum,FBS)诱导的血管壁损伤的影响;MTT法检测RSV对VSMCs增殖的影响;q PCR法检测RSV对TRPC1 m RNA表达的影响;Western Blot法检测RSV对TRPC1蛋白表达的影响。结果与模型组比较,RSV在20200μmol·L-1范围内,可改善高血清诱导的血管壁损伤和管壁增厚现象;RSV可降低FBS诱导的VSMCs增殖(P<0.01),以200μmol·L-1浓度最为显著,此作用可被TRPC1抑制剂SKF96365所阻断;RSV各浓度组的TRPC1 m RNA及蛋白相对表达量显著降低(P<0.01)。结论 RSV抑制VSMCs的增殖可能通过影响TRPC1靶点来实现。

【Abstract】 Objective To investigate whether resveratrol(RSV)interferes with the proliferation of vascular smoothmuscle cells by inhibiting transient receptor potential canonical-channel 1(TRPC1). Methods Vascular ringsculture method was applied to observe the effects of different concentrations of RSV on fetal bovine serum(FBS)-induced injury and thickening of the blood vessels;the effect of RSV on the proliferation of vascular smooth musclecells(VSMCs)was detected by MTT assay;quantitative real-time RT-PCR(q RT PCR)and Western Blot wereperformed to measure TRPC1 m RNA and protein levels. Results Compared with the model group, within theconcentration range of 20 μmol · L-1-200 μmol · L-1, RSV can improve the high serum induced injury andthickening of the vessel wall. RSV can inhibit proliferation of VSMCs induced by FBS(P < 0.01),with the besteffect at a concentration of 200 μmol · L-1, which can be blocked by TRPC1 inhibitor SK96365. The relativeexpression levels of TRPC1 m RNA and protein were significantly decreased in all RSV groups(P < 0.01).Conclusion The inhibition effect of RSV on vascular smooth cell proliferation was achieved by affecting the TRPC1.
