两种新型功能化自组装多肽水凝胶的制备和体外研究

【摘要】 目的研究自组装多肽水凝胶的表征及其生物相容性、促轴突再生的作用。方法本文制备两种分别模拟神经生长因子(NGF)和脑源性神经营养因子(BDNF)的功能化自组装多肽,通过圆二色谱分析、扫描电镜和原子力显微镜观察其表征,并将其用于培养雪旺细胞和PC12细胞。结果扫描电镜结果表明这种纳米纤维水凝胶具有类似细胞外基质的三维网络结构,圆二色谱分析和原子力显微镜结果显示该水凝胶具有典型的β折叠的色谱。体外结果表明雪旺细胞能很好地在多肽水凝胶中生长、铺展,同时PC12细胞能在多肽水凝胶中长出不同长度的轴突。结论自组装多肽水凝胶具有类似细胞外基质的纳米纤维结构,与神经类细胞的生物相容性良好,并且能够促进轴突生长。

【Abstract】 Objective To identify the characterization of self-assembled peptide hydrogel, its biocompatibility, and its promotion of the role of axonal regrowth. Methods In this study, we prepared a three-dimensional functionalized self-assembling peptide nanofiber hydrogel containing two neurotropic peptides, CTDIKGKCTGACDGKQC and RGIDKRHWNSQ derived from NGF and BDNF, respectively, and observed its characterization through circular dichroism spectroscopy, scanning electron microscopy and atomic force microscopy. In vitro, the material was used to cultivation of schwann cells and PC12 cells. Results The results from scanning electron microscopy revealed a three-dimensional porous matrix within the hydrogel. The data from circular dichroism spectroscopy and atomic force microscopy confirmed that the peptides displayed β-sheet structure and self-assembled into long nanofibers. In vitro, Schwann cells growed well and spreaded out in the peptide hydrogel, besides the hydrogel could promote the neurite outgrowth of PC12 cells. Conclusion The self-assembled peptide hydrogel has a nanofiber structure similar to the extracellular matrix, and it exhibits good cell compatibility. Furthermore, the hydrogel containing CTDIKGKCTGACDGKQC and RGIDKRHWNSQ can promote the neurite outgrowth of PC12 cells.
