九节茶药材中鞣质含量测定方法的改进
Improved Method for the Determination of Tannins in Sarcandra Glabra( Thunb. ) Nakai
【摘要】 目的:对九节茶药材中鞣质含量的紫外分光光度测定方法加以改进。方法:对中国药典鞣质含量测定方法(干酪素法)进行改进,供试品溶液的制备采用加热回流法或加热超声法代替药典中的静置过夜法,并将干酪素粒度明确为极细粉,由此建立了九节茶药材中的鞣质含量测定方法。结果:方法改进后,没食子酸在1.002~10.016μg·ml-1范围内与其吸光度呈良好的线性关系(r=0.999 4),九节茶药材鞣质平均回收率为97.81%,RSD为1.89%(n=6)。结论:该测定方法解决了原方法中重复性差、准确度低、操作用时长的问题,提高了测定结果的准确度和检测效率。
【Abstract】 Objective: To improve the ultraviolet spectrophotometric method for the content determination of tannins in Sarcandra glabra( Thunb.) Nakai. Methods: The method for the content determination of tannins in Chinese Pharmacopoeia( casein method)was improved. Staying overnight for the preparation of the sample solution was replaced by reflux heating or ultrasonic heating and the particle of casein was superfine powder,and the determination method of tannins in Sarcandra glabra( Thunb.) Nakai was established. Results: Through the methodological investigation on the improved method,the concentration of gallic acid showed a good linear relationship with the absorbance within the range of 1. 002-10. 016 μg·ml-1( r = 0. 999 4). The average recovery of tannins in Sarcandra glabra( Thunb.) Nakai was 97. 81%,and the RSD was 1. 89%( n = 6). Conclusion: The method solves such problems as poor repeatability,low accuracy and long operation time of the original method,which improves the accuracy of the determination results and the detection efficiency.
