麻荆止咳颗粒对咳嗽变异性哮喘豚鼠凋亡抑制基因Bcl-2表达的影响

【摘要】 目的:观察麻荆止咳颗粒对咳嗽变异性哮喘豚鼠肺组织中嗜酸粒细胞上凋亡抑制基因Bcl-2m RNA及蛋白表达的影响,探讨该颗粒治疗CVA豚鼠可能的作用机制。方法:用Muraki法建立咳嗽变异性哮喘模型,随机分为模型组、治疗组,用RT-PCR法检测治疗后豚鼠右肺中叶支气管肺组织中Bcl-2m RNA水平,用免疫组化染色法测定Bcl-2蛋白的表达。结果:治疗组肺组织均浆中的Bcl-2 mRNA浓度及阳性细胞免疫组化评分明显低于模型组,有显著性差异(P <0.05、P <0.01)。结论:麻荆止咳颗粒通过影响咳嗽变异性哮喘豚鼠肺组织中Bcl-2m RNA及其蛋白表达,达到促进肺组织EOS凋亡,减轻气道炎症,从而发挥平喘作用。

【Abstract】 Objective: To observe the effect of Majing Zhike granule( 麻荆止咳颗粒) on the expression of apoptosis suppressor gene Bcl-2 m RNA and protein on the eosinophils in the lung tissue of cough variant asthmatic guinea pigs, and to explore the possible mechanism of this granule in the treatment of CVA guinea pigs. Methods: The model of cough variant asthma was established by Muraki method. They were randomly divided into the model group and the treatment group. The level of Bcl-2 mRNA in the middle lobe and bronchopulmonary tissues of the right lung of guinea pigs after treatment was detected by RT-PCR. The expression of Bcl-2 protein was measured by immunohistochemical staining. Results: The concentration of Bcl-2 mRNA in the lung homogenate of the treatment group and the immuno histochemical score of the positive cells were significantly lower than those in the model group(P<0.05, P<0.01). Conclusion: Majing Zhike granule( 麻荆止咳颗粒) could improve the expression of Bcl-2 mRNA and its protein in the lung tissue of cough variant asthma guinea pigs, so as to promote the apoptosis of EOS in lung tissue and reduce airway inflammation, thus exerting antiasthmatic effect.
