电针对功能性消化不良大鼠MEK-ERK1/2信号通路的影响
Influence of Electro-acupuncture on MEK-ERK1/2 Signaling Pathway in Functional Dyspepsia Rats
【摘要】 目的观察电针对功能性消化不良(FD)大鼠胃组织MEK-ERK1/2信号通路的影响。方法将40只大鼠随机分为空白组及造模组,其中空白组10只。对除空白组外的30只大鼠进行造模,将造模成功的20只大鼠再随机分为模型组及电针组各10只。造模方法均采用夹尾刺激配合不规则饮食的复合造模方法制备FD大鼠模型。造模后,电针组进行电针足三里干预10 d,干预结束后用WB对大鼠胃组织丝裂原活化蛋白激酶(MEK)、细胞信号调节激酶1/2(ERK1/2)、磷酸化细胞外信号调节激酶(p-MEK)、磷酸化细胞信号调节激酶1/2(p-ERK1/2)蛋白水平进行检测。结果与空白组比较,模型组大鼠胃组织MEK、ERK1/2、p MEK、pERK1/2蛋白表达水平升高(P <0.05);与模型组相比,电针组MEK、ERK1/2、p-MEK、p-ERK1/2蛋白表达水平下降(P <0.05)。结论电针干预可下调FD模型大鼠MEK、ERK1/2、pMEK、p ERK1/2蛋白表达水平。
【Abstract】 Objective: To observe the effects of electroacupuncture(EA) on MEK-ERK1/2 signaling pathway in functional dyspepsia rats. Methods: 40 rats were randomized into a the blank group(10 cases) and the model group(30 cases). 20 rats successfully modeled were randomly divided into the model group and EA group,10 rats in each. After successful modeling,EA group received EA for 10 days. At the end of treatment,protein expression of MEK,ERK1/2,pMEK and pERK1/2 was detected with WB. Results: Compared with the blank group,the protein expression of MEK,ERK1/2,pMEK and pERK1/2 in the stomach tissue increased in the model group(P <0.05). Compared with the model group,the protein expression of MEK,ERK1/2,pMEK and pERK1/2 decreased in the EA group(P < 0.05). Conclusion: EA can down-regulate the protein expression of MEK,ERK1/2,pMEK and pERK1/2 in rats with functional dyspepsia.
