醋酸铅染毒致人外周血淋巴细胞遗传损伤的研究
Study of lead acetate induced genetic damage of human peripheral blood lymphocytes
【摘要】 目的研究醋酸铅染毒致人外周血淋巴细胞遗传损伤情况。方法不同浓度醋酸铅染毒人外周血淋巴细胞,CCK-8试验检测细胞活力;染色体畸变试验检测醋酸铅染毒对人外周血淋巴细胞染色体损伤作用;免疫荧光染色检测DNA双链断裂损伤生物标志物组蛋白H2AX的磷酸化(γ-H2AX)表达情况。结果醋酸铅染毒抑制人外周血淋巴细胞活力,其24 h的IC50值为168.5μmol/L;醋酸铅染毒后,人外周血淋巴细胞染色体损伤程度和γ-H2AX焦点表达水平随醋酸铅浓度的增加和染毒时间的延长而升高,呈现明显的剂量效应和时间效应。结论醋酸铅对人外周血淋巴细胞有潜在的细胞毒性和遗传毒性。
【Abstract】 [Objective]To study the genetic damage of human peripheral blood lymphocytes exposed to lead acetate.[Methods]The human peripheral blood lymphocytes were exposed to different concentrations of lead acetate. The cell viability was detected by CCK-8 assay, and chromosome damage was detected by chromosome aberration test. The expression of γ-H2 AX, which is the DNA double strand break biomarker, was detected by immunofluorescence staining.[Results]Results After exposed to different concentrations of lead acetate, the activity of human peripheral blood lymphocytes was inhibited, and the IC50 value was 168.5 μmol/L at 24 h. The degree of chromosome damage and the expression level of γ-H2 AX focus in human peripheral blood lymphocytes increased with the increase of lead acetate concentration and the prolongation of exposure time, showing a significant dose effect and time effect.[Conclusion]Lead acetate has potential cytotoxicity and genotoxicity to human peripheral blood lymphocytes.
