高效氯氰菊酯对大鼠黑质小胶质细胞活化的影响
Effect of β-cypermethrin on microglial activation in substantia nigra of rats
【摘要】 目的通过亚急性毒性实验探讨高效氯氰菊酯对大鼠的神经毒性及其作用机制。方法将刚断乳的雄性Wistar大鼠按照体重随机分为高效氯氰菊酯高剂量组(60 mg/kg)、中剂量组(40 mg/kg)、低剂量组(20 mg/kg)和空白对照组(植物油),经口灌胃染毒30天。观察大鼠的一般状况,计算体重、食物利用率、脏体比的变化。病理组织学HE染色观察脑组织的形态学改变。免疫组织化学染色观察黑质部位多巴胺能神经元的损伤及小胶质细胞的活化状况。结果高效氯氰菊酯能够使大鼠体重降低,实验第4周体重高剂量组为(288.18±19.06)g,中剂量组为(292.10±26.00)g,低剂量组为(307.41±15.73)g,对照组为(313.92±10.29)g,染毒组与对照组相比,差异有统计学意义(P<0.01);脑体比升高,高剂量组为(0.70±0.07)%,中剂量组为(0.68±0.08)%,低剂量组为(0.63±0.04)%,对照组为(0.59±0.11)%,染毒组高于对照组且差异有统计学意义(P<0.01);对照组和低剂量组大鼠脑组织细胞形态正常,细胞核完整且染色均一;中剂量组可见细胞核固缩、碎裂、染色不均,胞体变小变圆;高剂量组变性细胞数量明显增多,胞浆染色加深;与对照组相比,同种异体移植炎性因子(AIF-1)阳性细胞高剂量组较对照组数量增加,染色加深,胞体增大呈巨噬细胞样,分支明显增粗。结论高效氯氰菊酯对大鼠具有神经毒性,小胶质细胞的活化可能是其毒作用的机制之一。
【Abstract】 [Objective]To investigate the neurotoxicity of beta-cypermethrin and its mechanism in rats by subacute toxicity test.[Methods] Newly weaned male Wistar rats were randomly divided into the cypermethrin high dose group(60 mg/kg),middle dose group(40 mg/kg),low dose group(20 mg/kg) and blank control group(vegetable oil) according to body weight,and treated by gavage for 30 days.The general condition of rats was observed and the weight,food utilization rate and organ coefficient were calculated.The morphological changes of brain tissue were observed with HE staining.The damage of dopaminergic neurons in substantia nigra and the activation of microglia were observed by immunohistochemical staining.[Results] β-cypermethrin can make rats’ weight decreased,high dose group(288.18±19.06) g,middle dose group(292.10±26.00) g,low dose group(307.41±15.73) g,blank control group(313.92 ±10.29) g,with significant difference between dose groups and control group(P <0.01).Brain-body-ratio increased,high dose group(0.70±0.07)%,middle dose group(0.68 ±0.08)%,low dose group(0.63 ±0.04)%,control group(0.59±0.11)%,with significantly high value in dose groups,compared with control group(P<0.01).In control group and low dose group,brain cells were normal in morphology,with intact nuclei and uniform staining.In the middle dose group,nuclei condensed,fragmented,dyed uneven,and cell bodies became smaller and round.In high dose group,the number of denatured cells increased significantly,and cytoplasm staining deepened.Compared with the control group,AIF-1 positive cells in high dose group were dyed deeper and the cell body increased like macrophage,its branch were significantly thickening.[Conclusion] β-cypermethrin has neurotoxic effects on rats.Microglia activation may be one of the toxic mechanisms.
