【摘要】 目的:探析通络治痹汤对关节软骨修复及生理功能恢复的影响。方法:成年新西兰白兔24只,按随机数字表法分为假手术组(A组)、模型组(B组)、藤黄健骨丸组(C组)和通络治痹汤组(D组)。通过改良Hulth法构建兔膝骨关节炎模型,分组喂养及药物灌胃8周后,收集兔血清、关节液及右侧膝关节。采用Lequesne MG膝关节评估级别评测实验兔活动能力,通过大体观察及HE染色观察膝关节软骨形态学改变,ELISA法检测IL-1β、TNF-α水平。结果:与A组对比,B、C、D组Lequesne MG评分、Mankin’s评分及IL-1β、TNF-α水平均显著升高;与B组比较,C组与D组的Lequesne MG评分及Mankin’s评分明显降低,大体观察及HE染色可见软骨表面更加光滑、软骨细胞排列更加有序等,IL-1β、TNF-α水平明显下降。结论:通络治痹汤可促进软骨修复,改善关节生理功能,其作用机制可能与修复软骨、抑制炎症反应有关。更多还原

【Abstract】 Objective: Investigate the effects of Tongluo Zhibi Decoction on cartilage repair and physiological function recovery. Methods: 24 New Zealand white rabbits were randomly divided into four groups: Sham-operation group( Group A),Model group( Group B),Tenghuang Jiangu pills group( Group C) and Tongluo Zhibi Decoction group( Group D).The modified Hulth method was employed to establish rabbit knee osteoarthritis model. Different groups of rabbits were feed routinely and administrate drugs intragastrically,respectively and then serum and right knee joints were collected for the next analysis. Lequesne MG knee joint evaluation was used to assess movement ability; general observation and HE staining was applied to investigate morphological changes; ELISA method was employed to determine the levels of IL-1β and TNF-α. Results: Compared with group A,there were significant increases of Lequesne MG score,Mankin’s score,and the levels of IL-1β and TNF-α. Lequesne MG score and Mankin’s score of group C and D markedly decreased,meanwhile,general observation and HE staining showed more smooth surface of cartilage,more ordered arrangement of chondrocyte and more clear tidal line,while IL-1β and TNF-α levels decreased,in contrast with group B. Conclusion: Tongluo Zhibi Decoction could enhance cartilage repair and improve the physiological function of knee joint involving repairing cartilage and inhibiting inflammatory response.更多还原
