芹黄素对氧化应激诱导的HaCat细胞XBP1表达的影响

【摘要】 目的研究芹黄素对过氧化氢诱导的HaCat细胞氧化应激及XBP1表达的影响。方法采用MTT法测定芹黄素对HaCat细胞的活力的影响,DCFH-DA法流式细胞术检测HaCat细胞活性氧(ROS)含量,实时定量PCR检测HaCat细胞XBP1的mRNA表达。结果 500μmol/L的过氧化氢可显著诱导HaCat细胞ROS含量及XBP1s mRNA表达升高(P<0.01);10μmol/L及25μmol/L的芹黄素可显著抑制过氧化氢引起的HaCat细胞ROS含量及XBP1s mRNA表达升高,差异有统计学意义(P<0.01)。而过氧化氢或芹黄素对HaCat细胞未剪切的XBP1(XBP1u)的mRNA水平无明显影响。结论芹黄素可能通过抗氧化,对过氧化氢引起的XBP1s表达升高具有抑制作用,因而可能具有开发成为治疗白癜风的新药的前景。

【Abstract】 Objective In order to investigate the effects of apigenin on oxidative stress-induced expression of X-box binding protein 1(XBP1) in HaCat cells. Methods Effects of apigenin on cell viability were measured by MTT assay. Reactive oxygen species(ROS) generation was detected by cytometry analysis with DCFH-DA method,while m RNA levels of XBP1 were measured by reverse transcription quantitative PCR. Results Treatment with 500 μmol/L H2O2 induced significant increase of ROS levels and spliced XBP1(XBP1s) mRNA levels. Pretreatment with 10 μmol/L and 25 μmol/L apigenin significantly inhibited H2O2-induced increase of ROS levels and m RNA expression of XBP1 s,while mRNA levels of unspliced XBP1(XBP1u) were not significantly influenced by H2O2 or apigenin. Conclusion Apigenin decreased H2O2-induced ROS generation and expression of XBP1 s,thus may have a potential use in treating vitiligo.
