活动性结核患者外周血单核细胞Toll样受体2表达及其意义
Significance and expression of Toll-like receptor 2 in peripheral blood monocytes from active tuberculosis patients
【摘要】 目的通过检测活动性结核(TB)患者外周血单核细胞Toll样受体2(TLR2)表达及其活化后单核细胞功能的变化,探讨单核细胞TLR2样受体在机体感染结核过程中的作用。方法收集32例活动性TB患者和32例健康对照者外周血,流式细胞术检测单核细胞TLR2表达;体外分离培养健康人外周血单核细胞,通过脂阿拉伯甘露糖(LAM)活化TLR2,流式细胞术检测LAM诱导的巨噬细胞比例及细胞凋亡。结果与对照组相比,TB患者外周血TLR2阳性细胞比例和平均荧光强度均显著上升(P <0. 05)。体外分离培养健康人外周血单核细胞,与对照组相比,LAM诱导的巨噬细胞CD86阳性细胞比例和凋亡细胞比例均增加(P <0. 05),抗TLR2封闭型抗体可抑制LAM诱导的巨噬细胞分化和凋亡。结论单核细胞TLR2在结核疾病过程中发挥重要的作用。
【Abstract】 Objective To explore the role of Toll-like receptor2( TLR2) in peripheral blood monocytes in the pathogenesis of tuberculosis,this paper investigated the expression of TLR2 in patients with active tuberculosis( TB) and the function of monocytes after TLR2 activation. Methods After the peripheral blood of 32 TB patients and 32 healthy controls were collected,the expressions of TLR2 on monocyte were detected by flow cytometry. After monocytes were isolated from healthy people,TLR2 was activated by lipoarabinomannan( LAM),and the expression of CD86 and apoptosis of macrophages were measured by flow cytometry. Results The percentage of TLR2 positive cells on peripheral blood monocytes of TB patients were higher than that of the control group( P < 0. 05),as well as the TLR2 mean fluorescence intensity. After the monocytes were isolated from healthy people,the percentage of CD86 positive macrophages cells and apoptotic cells induced by LAM increased significantly,compared to the control group( P < 0. 05). However,LAM-induced macrophage differentiation and apoptosis were inhibited by anti-TLR2 blocking antibody. Conclusion TLR2 in monocytes may play an important role in the pathogenesis of tuberculosis.
