福州市柯萨奇A6型流行情况及VP1区基因特征分析
Epidemic situation and gene characteristics analysis for VP1 region of Coxsackie A6 in Fuzhou
【摘要】 目的了解福州市柯萨奇病毒A6型(Cox A6)的流行情况及VP1区基因特征及种系进化分布。方法采集临床诊断为手足口病患者的咽试子或肛拭子标本,通过real-time PCR法选取部分重症患者柯萨奇病毒A6型阳性标本,采用RD细胞分离病毒,RT-PCR扩增VP1基因片段并测序,利用DNAStar和Mega 5. 1软件进行核苷酸和氨基酸同源性分析并构建亲缘进化树。结果检出的3 237份肠道病毒阳性样本中,肠道病毒71型(EV71)、柯萨奇病毒A组16型(Cox A16)、Cox A6的阳性率分别为22. 0%、14. 4%、23. 9%,不同肠道病毒阳性率差异有统计学意义(P <0. 05)。福州市CVA6型分离株的核苷酸和氨基酸同源性分别为96. 3%~100%和97. 4%~100%,与原型株CV-A6/Gdula的同源性分别为81. 6%~83. 5%和94. 1%~95. 7%。进化树的分析结果显示,福州市Cox A6分离株属于D基因型。结论 Cox A6是引起福州市手足口病的主要病原体之一,本次分离得到的Cox A6病毒株均属于D基因型。
【Abstract】 Objective To understand the epidemic characteristics of VP1 gene and evolutionary distribution of Coxsackie A6 virus in Fuzhou. Methods Throat or anal swabs were collected from suspected hand-foot-mouth disease( HFMD) patients,and positive specimens of Coxsackie A6 virus were selected by real-time PCR. Viruses were isolated with RD cells. VP1 genes of Coxsackie A6 viruses were amplified by RT-PCR. Homological analysis of nucleotide and amino acid was performed and phylogenetic tree was constructed with DNAStar MegAlign and Mega 5. 1 software. Results A total of 3 237 positive samples were detected,and the positive rates of EV71,Coxsackie A16 and Coxsackie A6 were 22. 0%,14. 4%,23. 9%,respectively.The difference in positive rates of different intestinal virus was statistically significant( P < 0. 05). The homology of nucleotide and amino acid among the Cox A6 isolated strains were within 96. 3%-100% and 97. 4%-100%,respectively. Compared with the Cox A6 prototype strain( Gdula strain),the homology of nucleotide and amino acid was within 81. 6%-83. 5% and94. 1%-95. 7%,respectively. Phylogenetic tree showed that Cox A6 isolated strains were most similar to the gene subtype D.Conclusion Coxsackie A6 is one of the major pathogens causing HFMD in Fuzhou. Evolution analysis shows that the Cox A6 strains isolated in Fuzhou belongs to a genotype D.
