辽宁省2014年-2016年季节性A(H3N2)流感病毒耐药基因分析
Analysis of drug resistance of influenza A(H3N2) virus in Liaoning province during 2014-2016
【摘要】 目的分析辽宁省2014年-2016年甲型H3N2亚型流感毒株基质蛋白2(matrix protein2,M2)和神经氨酸酶(neuraminidase,NA)基因特点,掌握辽宁省内流感流行株对烷胺类药物以及神经氨酸酶抑制剂类药物的耐药情况,为流感的临床治疗及预防控制提供依据。方法选取甲型H3N2亚型流感毒株51株,提取核酸后对M2基因和NA基因进行PCR扩增,测序后对耐药位点进行分析。结果所选毒株均发生了M2基因的S31N耐药位点突变,M2基因的其余耐药位点和NA基因的全部耐药位点未发现耐药突变。结论辽宁省2014年-2016年流行的甲型H3N2型流感毒株普遍对烷胺类药物耐药,对神经氨酸酶抑制剂类药物敏感。奥司他韦、扎那米韦等NA抑制剂类药物依然是近年来甲型H3N2流感防治的敏感药物。
【Abstract】 Objective Analyze the characteristics of matrix protein 2( matrix,protein2,M2) and neuraminidase( Neuraminidase,NA) gene of influenza A( H3 N2) subtype in Liaoning Province during 2014-2016,and master the resistance of influenza pandemic strains to amantadine and neuraminidase inhibitors,so as to provide basis for clinical treatment and prevention and control of influenza. Methods 51 strains of influenza A( H3 N2) were selected. After nucleic acid extraction,PCR amplification was conducted on M2 gene and NA gene,and after sequence,resistance loci were analyzed. Results S31 N mutations of M2 were found in the selected strains,and no mutated site was found in neuraminidase inhibitors. Conclusion Influenza A( H3 N2) strains prevalent in Liaoning province during 2014-2016 are generally resistant to amantadine and are sensitive to neuraminidase inhibitors. Oseltamivir and zanamivir are still the sensitive drugs for influenza A H3 N2 influenza prevention and treatment in recent years.
