貉细小病毒SD1607株的分离鉴定及VP 2基因序列分析

【摘要】 从疑似患细小病毒性肠炎的貉子肠道组织样品中分离到一株貉细小病毒(RDPV-SD1607)。为研究其分子遗传特征,对该分离株VP2基因进行克隆序列分析,结果显示,RDPV-SD1607的VP2序列与RDPV-HB3的相似性最高,为99.49%,属于CPV-2亚型;利用其核苷酸序列建立系统进化树发现,RDPV-SD1607处于犬细小病毒分支,且处于CPV-2亚型和CPV-2a亚型分支之间。结果表明,RDPV-SD1607株与犬细小具有共同的遗传起源,推测其可能正处于CPV-2亚型向CPV-2a亚型进化的中间状态,或是CPV适应貉而形成的新毒株。

【Abstract】 A virus named RDPV-SD1607 was isolated from the intestinal tissue of a diarrheal raccoon dog. To analyze the molecular and genetic characteristics of parvovirus originated from raccoon dog,the nucleotide sequence analysis of VP2 gene indicated that the isolate strain belonged to CPV-2 strain,and shared the highest similarity( 99. 49%) with RDPV-HB3. The phylogenetic tree analysis showed that the RDPV-SD1607 was located in the same branch with the canine parvovirus,but in the different branch with feline parvovirus. Besides,it was situated between the CPV-2 and CPV-2 a. This study suggests that the RDPV-SD1607 and canine parvovirus might originates from the same genetic ancestors,and it might be the evolutionary strain from CPV-2 to CPV-2 a,or a new virus strain which adapts to the new host( raccoon dog).
