高活性云芝α-半乳糖苷酶发酵培养基的优化

【摘要】 研究了云芝(Coriolus versicolor)液体发酵过程中不同诱导物及培养基组成对α-半乳糖苷酶活性的影响。采用L9(34)正交设计法对云芝发酵培养基进行了优化,确定了其发酵产酶的最佳培养基。结果表明,云芝发酵生产α-半乳糖苷酶的最佳诱导物是豆粕粉,最优培养基为豆粕粉2%、半乳糖0.5%、KH2PO4 0.1%、MgSO4 0.1%,发酵培养8 d,酶活性为301.6 U·mL-1

【Abstract】 To determine the optimum culture medium for the α-galactosidase production during the fermentation of Coriolus versicolor, the effects of different inducer and medium composition on the activity of α-galactosidase had been studied. The results of L9(34) orthogonal experiment showed that the best inducer for α-galactosidase production was soybean meal, and the optimum medium composition was soybean meal 2%, galactose 0.5%, KH2PO4 0.1%, MgSO4 0.1%, and the fermentation time was 8 days, finally the enzyme activity could reach 301.6 U·mL-1.
