应用过氧化物同工酶鉴定山西省马铃薯主栽品种
Identification of Main Potato Varieties in Shanxi with Peroxidase Isozyme
【摘要】 为快速、准确鉴定马铃薯不同品种。本研究以14个马铃薯品种的芽部、茎部、叶片为实验材料,分别提取其过氧化物同工酶,并进行聚丙烯酰胺凝胶电泳。通过比较不同品种马铃薯同工酶的酶谱差异,来确定马铃薯之间亲缘关系的远近。结果表明,马铃薯植株芽的过氧化物同工酶可以作为区分马铃薯品种的一个重要指标,14个品种中4、7号品种酶带相似,分别为‘晋薯11号’和‘晋薯14号’;6、9号品种酶带相似,分别为‘晋薯21号’和‘晋薯18号’;5、12号品种酶带相似,分别为‘同薯22号’和‘同薯20号’;13、14号品种酶带相似,品种分别为‘05-32-7’和‘同薯23号’;表明这些品种之间亲缘关系较近。
【Abstract】 To identify the varieties of potato quickly and accurately, peroxidase isoenzyme was extracted fromthe buds, stems and leaves of 14 potato varieties, and separated with polyacrylamide gel electrophoresis. Thedifferences of zymogram were compared to confirm their genetic relationships. The results showed thatperoxidase isoenzyme of buds could be used as an important criterion for distinguishing potato varieties.Within the 14 varieties, the enzyme bands of No.4 and No.7 were similar, which represented‘Jinshu 11’and‘Jinshu 14’, respectively. The enzyme bands of No.6 and No.9 were similar, which corresponded to‘Jinshu21’and‘Jinshu 18’, respectively. The enzyme bands of No.5 and No.12 were similar, which represented‘Tongshu 22’and‘Tongshu 20’, respectively, and the enzyme bands of No.13 and No.14 were similar, whichcorresponded to‘05-32-7’and‘Tongshu 23’. These results indicated that the genetic relationships ofcultivars with similar enzyme bands were closer.
