单因素结合响应面法优化蒙香青兰总酚含量测定方法
Single Factor Combined with Response Surface Methodology to Optimize the Determination Method of Total Phenol Content in Mongolia Dracocephalum Moldavica L.
【摘要】 目的:以蒙香青兰为研究对象,优化建立福林酚比色法测定总酚含量的测定方法。方法:先利用单因素实验确定测定方法的主要影响因素,再采用Box-Behnken响应面法对检测条件进行优化。结果:取适量蒙香青兰总酚提取物,加入3. 95mL去离子水漩涡混匀后,再加入0. 6mL福林酚试剂,漩涡混匀并避光静置5min。之后加入20%碳酸钠1. 2mL,漩涡混匀后加去离子水定容至10mL,摇匀,于55℃恒温水浴箱中避光反应30min,在760nm波长测吸光度。结论:该测定方法操作简便快捷,设备要求低,重复性及重现性良好,且50min内测定结果稳定。
【Abstract】 To optimize Folin-Ciocalteu( FC) colorimetric determination of total phenol content in Mongolia Dracocephalum Moldavica L. On the basis of single factor experiment,the detection conditions of total phenol from Mongolia Dracocephalum Moldavica L. were further optimized by Box-Behnken design. The results showed the optimal condition for detection was as follows: 3. 95 mL deionized water was added into appropriate amount of total phenol from Mongolia Dracocephalum Moldavica L.,after vortex mixing,adding 0. 6 mL FC,then placing in darkness and reacting for 5 min. Then,1. 2 mL of 20% Na2 CO3 was added,and mixed with deionized water,then placing in darkness and reacting for 30 min in 55 ℃,finally measuring absorbance at the wavelength of 760 nm. The developed method had good repeatability and reproducibility,as well as high stability within 50 min.
