不同流行区间日疟原虫疫苗候选抗原pvama1基因DⅠ结构域的种群结构分析
Global population structure analysis of DⅠ domain of Plasmodium vivax vaccine candidate pvama1
【摘要】 目的:明确中缅边境流行区间日疟原虫(P.vivax) pvama1基因DomainⅠ(DⅠ)区域的多态性和种群结构。方法:提取16例中缅边境P. vivax感染患者血gDNA; PCR和测序检测其pvama1基因DⅠ区域,参考韩国、泰国和巴布新几内亚(PNG)流行区相应序列,采用Dna SP6. 10. 1和Structure软件进行多态性、中性检验和种群结构分析。结果:成功扩增16例中缅边境pvama1基因DⅠ区;各流行区pvama1基因的DⅠ区π值为0. 013~0. 021,且d_N/d_S>1(P<0. 05)。PNG流行区Tajima’s D~*值显著大于1(P<0. 05);各流行区pvama1基因分为3个亚群(K=3)。结论:Pvama1基因DⅠ区具有低多态性并经历阳性选择,提示其为红内期疫苗候选靶位;中缅边境、泰国和PNG流行区具有相似的种群结构,提示以其主要单倍体型为基础设计的疫苗具有广泛的保护作用。
【Abstract】 Objective: To determine the genetic diversity and population structure of DomainⅠ(DⅠ) of Plasmodium vivax(P. vivax) apical membrane antigen 1(pvama1) in Yunnan province China-Myanmar border area. Methods: Genomic DNA was extracted from 16 P. vivax infected patients’ blood samples in China-Myanmar border area. The Domain I(DⅠ) of pvama1 gene was amplified by PCR and sequenced. pvama1 DⅠ sequence of Korea,Thailand and PNG was achieved from Gen Bank database. Genetic diversity,neutral tests and population structure analysis were performed by Dna SP6. 10. 1 and STRUCTURE software. Results: The DⅠ domain of pvama1 was successfully amplified from 16 samples. A π value ranging from 0. 013 to 0. 021 and d_N/d_S>1(P<0. 05) was detected in all studied endemic areas. The value of Tajima’s D~* was significantly over 1 in PNG(P<0. 05). Population structure analysis indicated that the haplotypes were optimally grouped into 3 clusters(K = 3). Conclusion: The pvama1 DⅠ domain sequences from endemic areas show limited genetic diversity and was under positive diversifying selection. These results indicates that DⅠ domain is a potential vaccine target region for blood stage parasites. China-Myanmar border,Thailand and PNG endemic areas showed similar clustering patterns,indicating candidate pvama1 vaccine based on these antigenically similar haplotypes may have extensive protecting efficiency.
