IL-17A在乳腺浸润性导管癌中表达及对MCF-7细胞增殖转移的作用

【摘要】 目的考察IL-17A在乳腺癌(BIDC)和乳腺纤维腺瘤(FAB)中的表达及其对MCF-7细胞体外增殖转移的作用,探讨IL-17A在乳腺癌发生、发展中的意义及其作为乳腺癌基因治疗靶点的可能性.方法选取BIDC 50例及FAB 20例,免疫组化法检测IL-17A表达;与IL-17A共孵育后,MTT法和Transwell小室法检测MCF-7细胞体外增殖转移.结果 IL-17A在IDC中表达率((6.27±1.22)%)高于FAB((3.07±0.78)%),差异具有统计学意义(P<0.01);经不同浓度IL-17A共孵育后,MCF-7细胞增殖无明显差异(P>0.05);与100μg/L的IL-17A共孵育后,穿过Transwell小室膜的细胞数((84±8)/HP)高于对照组((32±6)/HP),差异具有统计学意义(P<0.01).结论 IL-17A在BIDC中的表达高于FAB,体外虽未促进MCF-7细胞的增殖,但能增强其侵袭能力,有望作为乳腺癌基因治疗的靶点.

【Abstract】 Objective By investigating expression of IL-17 A in breast infiltrating ductal carcinoma( BIDC) and fibroadenoma of breast( FAB),and its function in vitro on proliferation and metastasis of MCF-7 cells,to explore the significance of IL-17 A on breast cancer development and its potential as a gene therapeutic target for breast cancer. Method Fifty cases of BIDC and 20 cases of FAB were selected,expression of IL-17 A was investigated by immunohistochemistry; In vitro proliferation and metastasis of MCF-7 were detected by MTT and Transwell methods after co-incubation with IL-17 A. Results Expression of IL-17 A in BIDC was higher than that in FAB( 6. 27±1. 22) % vs( 3. 07±0. 78) %( P<0. 01); After co-incubation with IL-17 A,no significant difference of proliferation was found( P > 0. 05); but after co-incubation with 100 μg/L of IL-17 A,cells that through the membrane were higher than control group(( 84 ±8)/HP vs( 32 ±6)/HP,P<0. 01). Conclusion Expression of IL-17 A in BIDC was higher than that of FBA; IL-17 A could not promote MCF-7 proliferation in vitro,but enhance their abilities of metastasis; IL-17 A was a potential target for breast cancer gene therapy.
