去甲斑螯素复合人工骨的理化性质及体内置入实验
Physico-chemical properties of bioceramics composited with norcantharin and its in vivo test
【摘要】 [目的]测试复合去甲斑螯素(NCTD)的生物陶瓷理化性能并研究NCTD复合人工骨对成骨性能的影响和生物安全性。[方法]将10μg/ml的NCTD溶液,浸泡多孔生物陶瓷,并风干、冷冻干燥。分别对复合NCTD多孔生物陶瓷及普通生物陶瓷进行理化数据测量,包括气孔率、力学强度、钙磷比测定。将NCTD生物陶瓷置入骨缺损模型动物体内,取出标本进行组织学V-G染色,观察其对成骨性能的影响。定期检测动物的血常规、肝功能,评价置入物的生物毒性。[结果]复合NCTD后生物陶瓷,钙磷比由1.51变为1.49,力学强度由3.28 MPa变为3.20 MPa,空白与复合NCTD陶瓷上述指标差异均无统计学意义(P>0.05)。随置入时间延长,组织切片显示两组置入物残余明显减小,而新生骨量增加。在置入空白与复合NCTD生物陶瓷后,白细胞及血清谷丙转氨酶术前、术后1周、术后2周各时间点无明显变化,两组间差异无统计学意义(P>0.05)。[结论]复合NCTD后的多孔生物陶瓷理化性质较普通生物陶瓷无差异。具有良好的促骨生长能力,无生物毒性。
【Abstract】 [Objective] To test the physical and chemical properties of bioceramics composited with norcantharin(NCTD),and explore its effect on osteogenesis as well as its biological safety. [Methods] After immersed in the NCTD solution at a concentration of 10 ug/ml, the porous bioceramics implants were air-dried and lyophilized. The physical and chemical characters of blank porous bioceramics and that composited with NCTD were measured respectively, including porosity, mechanical strength,calcium and phosphorus ratio. After that, both blank bioceramics and that composited with NCTD were implanted into the femoral bone defect in rabbits. After 4, 8 and 12 weeks, the specimens were harvested for histological slice with Van Gieson’s picrofuchsin stain(V-G stain) to observe the effect on osteogenesis. In addition, blood test will be conducted regularly to assess the biological toxicity of the implants. [Results] After composited with NCTD, the ratio of calcium to phosphorus varied from 1.49 to1.51, while the mechanical strength changed from 3.28 MPa to 3.20 MPa, where were not statistically significant between the blank implant and the NCTD composited implant(P>0.05). The histological observation showed that the residual implants significantly decreased, while the new bone increased over time. Regarding to biotoxicity, the white blood cell count and alanine transaminase kept no significantly changed in both groups among the time points before operation, at 1 and 2 weeks postoperatively(P>0.05), and no significant difference between the two group was noticed at any corresponding time point(P>0.05).[Conclusion] There are no differences between the blank porous bioceramics and the bioceramics composited with NCTD on the aspects of the physical and chemical properties. Both have good potential of bone ingrowth and no biological toxicity.
