血液RNA制备对微小RNA分析结果的影响

【摘要】 血液微小RNA (miRNA)有望成为某些疾病诊断和治疗监测的生物学标志,因此对血液微小RNA灵敏、精确的定量分析成为检验学的重要研究内容。血液分析样本制备对灵敏、精确的定量分析至关重要。综述血液处理和样本制备对微小RNA分析结果的影响。Trizol法提取的总RNA样本可以获得更准确的miRNA分析结果;去除干扰组分的小分子RNA样本可以提高微阵列和实时荧光反转录PCR分析miRNA的灵敏度。此外,RNA样本质量、数量及血液收集时血样体积、抗凝剂选择、溶血发生都可以影响miRNA分析结果的灵敏度和准确性。

【Abstract】 Blood microRNAs is expected to become a biomarker for diagnosis and treatment monitoring of some diseases.Therefore,sensitive and accurate quantitative analysis of blood microRNAs become an important research field of laboratory medical science.Sample preparation for blood analysis is essential for sensitive and accurate quantitative analysis.Influence of blood processing and sample isolation on microRNAs analysis results was reviewed.Total RNA samples extracted by Trizol method can obtain more accurate results of microRNAs analysis.Small RNA samples removing interference substances may enhance the sensitivity of RT-dPCR and microarray-based microRNAs analysis.The sensitivity and accuracy in microRNAs assay is also influenced by the quality and the amount of RNA sample derived from the blood,blood volume,anticoagulant selection and hemolysis.
