谷氨酸功能异常与阿尔茨海默病
Glutamate Dysfunction and Alzheimer’s Disease
【摘要】 阿尔茨海默病(Alzheimer’s disease,AD)是全球最流行的痴呆症形式,是一种多因素参与的复杂的神经退行性疾病。研究表明谷氨酸功能异常参与了AD的发生和发展,谷氨酸兴奋性毒性是AD进展的重要原因之一。从认知功能正常发展至AD过程中,谷氨酸功能呈现波动性变化,这为以谷氨酸功能通路为靶点的干预措施的研究增加了难度。该文的目的是对谷氨酸功能异常与AD之间的关系进行综述,通过讨论谷氨酸的正常生理过程和功能,延伸到其病理过程,并针对谷氨酸通路中可能的药物靶点进行探究。
【Abstract】 Alzheimer’s disease(AD),which is the most prevalent form of dementia,is a multi-factorial induced complicated neurodegenerative disease. Glutamate dysfunction was involved in AD and glutamate excitotoxicity is one of the most important pathogenesis of AD.The function of glutamate fluctuates during the development from normal cognition to AD,which increase the difficulty of intervention study. The aim of this review is to discuss the relationship between glutamate dysfunction and AD. By discussing the physiological and pathological function of glutamate,we probe the possible drug targets for AD in glutamatergic pathway.
