【摘要】 目的探讨仙灵骨葆胶囊对去卵巢大鼠骨组织中雌激素受体α(estrogen receptorα,ERα)、组蛋白去甲基化酶(JMJD2B)、人Runt相关转录因子2(runt-related transcription factor 2,Runx2)、碱性磷酸酶(alkaline phosphatase,ALP)的表达水平。方法按随机数字表法将40只SD雌性大鼠分为4组:空白对照组、阴性对照组、实验组、阳性对照组,各10只。空白对照组大鼠保留双侧卵巢,仅切除卵巢旁部分脂肪,另外3组大鼠经手术切除双侧卵巢,制备骨质疏松模型。术后1周,实验组按30.85 mg/100 g比例给予仙灵骨葆胶囊,阳性对照组按11.7 mg/100 g比例给予福美加,空白对照组、阴性对照组喂养同体积蒸馏水。持续用药3个月后,采用逆转录-聚合酶链式反应(RT-PCR)检测大鼠股骨组织ERα、JMJD2B、Runx2和ALP mRNA表达,Western Blot检测ERα、JMJD2B蛋白表达。结果 与空白对照组相比,实验组大鼠股骨组织中ERα、JMJD2B蛋白及其mRNA、Runx2、ALP表达量明显下降,两组对比有统计学意义(P<0.05);与阴性对照组比较,实验组ERα、JMJD2B蛋白及其mRNA、Runx2、ALP表达量均明显上升,两组对比有统计学意义(P<0.05);与阳性对照组比较,实验组ERα、JMJD2B蛋白及其mRNA表达量明显上升,两组对比有统计学意义(P<0.05),而Runx2表达量明显下降(P<0.05)。结论 本实验结果发现仙灵骨葆胶囊可提高ERα、JMJD2B、Runx2、ALP的表达。更多还原

【Abstract】 Objective To investigate the effects of Xianlinggubao Capsule on the expression levels of Estrogen Receptor α(ERα), histone demethylase(JMJD2 B), and Runt-related transcription factor 2(Runx2) and Alkaline Phosphatase(ALP). Methods Forty SD female rats were divided into four groups according to the random number table: blank control group, negative control group, experimental group and positive control group, with 10 rats in each group. Rats in the blank control group retained bilateral ovaries and only excised paraesophageal fat. Rats in the other three groups were subjected to surgical resection of bilateral ovaries to prepare an osteoporotic model. One week after surgery, the experimental group was given Xianlinggubao Capsules at a ratio of 30.85 mg/100 g, the positive control group was given FuMeiJia at a ratio of 11.7 mg/100 g, and the blank control group and the negative control group were fed with the same volume of distilled water. After 3 months of continuous medication, RT-PCR was used to detect the mRNA expression of ERα, JMJD2 B, Runx2 and ALP in rat femur. Western Blot was used to detect the expression of ERα and JMJD2 B protein. Results Compared with the blank control group, the expression of ERα, JMJD2 B protein and its mRNA, Runx2 and ALP in the femur tissue of the experimental group decreased significantly, and there were statistically significant differences between the two groups(P<0.05). Compared with the negative control group, the expression of ERα, JMJD2 B protein and its mRNA, Runx2 and ALP in the experimental group increased significantly(P<0.05). Compared with the positive control group, the expression of ERα, JMJD2 B protein and its mRNA in the experimental group increased significantly, with statistically significant differences between the two groups(P<0.05), while the expression level of Runx2 decreased significantly(P<0.05). Conclusion The result of this experiment found that Xianlinggubao Capsule could increase the expression of ERα, JMJD2 B, Runx2 and ALP.更多还原
