红杉醇抑制葡萄糖苷酶及改善糖耐量实验研究

【摘要】 目的探讨红杉醇体外对葡萄糖苷酶的抑制作用以及对糖耐量的影响。方法采用96孔板法,分别测定红杉醇对α-葡萄糖苷酶、β-葡萄糖苷酶、α-半乳糖苷酶、β-半乳糖苷酶、α-淀粉酶、葡萄糖淀粉酶、蔗糖酶和海藻糖酶的抑制作用,以及抑制类型。对小鼠灌胃给予红杉醇后,考察其糖耐量。结果红杉醇等化合物对α-葡萄糖苷酶具有较强的抑制作用,对β-半乳糖苷酶,α-淀粉酶,葡萄糖淀粉酶和海藻糖酶也有一定的抑制作用,且抑制类型多为竞争性抑制。结论红杉醇在一定程度上具有降血糖的作用,其作用机制部分通过抑制-葡萄糖苷酶实现。红杉醇对糖尿病的预防与治疗具有一定作用。

【Abstract】 Objective To explore the inhibitory effect of sequoyitol on glucosidase activity in vitro and the effect on glucose tolerance in mice. Methods 96-well plate method was used to observe the inhibitory effect and the type of inhibition of sequoyitol onα-glucosidase,β-glucosidase,α-galactosidase,β-galactosidase,α-amylase,glucoamylase,sucrose and trehalase in vitro.In addition,the glucose tolerance was determined by intragastric administration of sequoyitol to mice. Results Sequoyitol could competitively inhibit the enzymatic effects ofα-glucosidase,β-galactosidase,α-amylase,glucoamylase,trehalase and the inhibitory effect was the strongest onα-glucosidase.Intragastric administration of sequoyitol in mice could also reduce the increase of postprandial blood glucose level. Conclusions Sequoyitol as an active ingredient can lower the blood sugar by inhibiting the activity of glucosidase,especiallyα-glucosidase,therefore it may be used to prevent and treat diabetes.
