河南省烟粉虱传播的番茄病毒病分子鉴定
Molecular identification of whitefly transmitted tomato viruses in Henan
【摘要】 为了探究河南省番茄主产区是否受到烟粉虱传播的番茄黄化曲叶病毒、番茄褪绿病毒和番茄侵染型褪绿病毒的复合侵染,采集叶片边缘卷曲、叶片皱缩、叶脉间黄化褪绿、植株矮化的疑似烟粉虱传播的3种病毒病的番茄植株样品,分别用TYLCV、ToCV和TICV特异性引物对样品进行PCR和RT-PCR检测。结果表明,TYLCV和ToCV的特异性引物分别扩增得到约780 bp和460 bp左右的特异性条带,该条带与TYLCV和ToCV阳性对照大小一致,且与这2种病毒郑州分离物核苷酸序列同源性均在99%以上;TICV特异性引物未扩增出目的条带。说明河南主栽区番茄受到烟粉虱传播的番茄褪绿病毒和番茄黄化曲叶病毒的复合侵染。
【Abstract】 In order to explore the viruses which transmitted by whitefly, Tomato chlorosis virus(ToCV), Tomato yellow leaf curl virus(TYLCV) and Tomato infectious chlorosis virus(TICV) were studied in this paper. The samples which showed symptoms of marginal curling, leaf shrinkage, interveinal chlorosis and dwarf plant were collected in the main tomato areas of Henan Province. Using specific primers of ToCV, TYLCV and TICV, ToCV, TYLCV and TICV were detected by RT-PCR and PCR. The result showed that, the specific primers of TYLCV and ToCV amplified a 780 bp specific band and a 460 bp specific band separately. The two specific bands were amplified with the same size as the positive control by the two sets of specific primers, and the nucleotide sequence homology between these two viruses and Zhengzhou isolates was above 99%. But no bands of TICV were amplified by RT-PCR by the set of specific primers.Tomato plants were infected by both ToCV and TYLCV in the main planting areas of Henan.
