清肝降压胶囊对实验性眼压升高的抑制作用
Qinggan Jiangya capsule inhibiting experimental intraocular pressure elevation
【摘要】 目的观察清肝降压胶囊对大鼠实验性眼压升高的影响。方法 Wister大鼠右眼前房注射乳化硅油制作眼压升高模型,左眼作为正常对照。造模后第2天将实验动物分为蒸馏水对照组、低剂量清肝降压胶囊组(20mg/ml),中剂量清肝降压胶囊组(40mg/ml),高剂量清肝降压胶囊组(80mg/ml),醋甲唑胺组(0.9 mg/ml),按1 ml/100 g体质量灌胃给药,每天给药1次,第15天停药。测定第3、5、8、12、15天时的眼压,观察眼压变化及组间差异。结果模型眼造模后眼压(7.46±1.88)mm Hg(1mmHg=0.133kPa),高于造模前的(6.28±1.42)mmHg(配对t检验,t=4.491,P<0.001)。对于模型眼,20 mg/ml清肝降压胶囊组在第5、12天,眼压低于蒸馏水对照组(P<0.05);40 mg/ml清肝降压胶囊组在第5、12天,眼压低于蒸馏水对照组(P<0.05);80 mg/ml清肝降压胶囊组在第3、5、8、12,眼压低于蒸馏水对照组(P<0.05);醋甲唑胺组在第3、5、8、12天,眼压低于蒸馏水对照组(P<0.05)。第15天,各处理组眼压数值均与蒸馏水对照组接近,组间差异无统计学意义(P>0.05)。对于正常眼,三个剂量的清肝降压胶囊组各时间点的眼压始终与蒸馏水对照组无明显差异(P>0.05);醋甲唑胺组在第8、12天,眼压低于蒸馏水对照组,差异有统计学意义(P<0.05),第15天停药后眼压与蒸馏水对照组接近(P>0.05)。结论清肝降压胶囊对实验性眼压升高有抑制作用,对正常眼眼压无明显影响。醋甲唑胺对实验性眼压升高和正常眼均有降眼压作用。
【Abstract】 OBJECTIVE To examine roles of inhibiting experimental intraocular pressure(IOP) elevation by Qinggan Jiangya capsule. METHODS 20 mg/ml, 40 mg/ml and 80 mg/ml Qinggan Jiangya capsule was used, distilled water as negative control and 0.9 mg/ml methazolamide as positive control. Right eyes of rats were modeled by intraocular pressure elevation. Left eyes were normal eyes. Treatment started on second day, as fifteenth day withdrawal. RESULTS On the third day, 80 mg/ml Qinggan Jiangya capsule and methazolamide group had significant inhibiting IOP elevation effect in experimental eyes.On the fifth day, 20 mg/ml, 40 mg/ml,80 mg/ml Qinggan Jiangya capsule and methazolamide group had signifi cant inhibiting IOP elevation effect. On the eighth day, 80 mg/ml Qinggan Jiangya capsule and methazolamide group had significant inhibiting IOP elevation effect. On the twelfth days, 20 mg/ml,40 mg/ml, 80 mg/ml Qinggan Jiangya capsule and methazolamide group had significant inhibiting IOP elevation effect. On the fifteenth days, there was no inhibiting IOP elevation effect in all groups. The methazolamide group had reducing IOP effect on the eighth and twelfth days in normal eyes. CONCLUSIONS Qinggan Jiangya capsule had inhibiting IOP elevation effect and no effect of reducing normal IOP. Methazolamide had inhibiting IOP elevation and reducing normal IOP effect.
