草地早熟禾查尔酮合成酶基因PpCHS1克隆、功能与表达分析

【摘要】 以草地早熟禾转录组数据为基础,克隆得到PpCHS1基因cDNA序列。其包含查尔酮合成酶基因(CHS)家族保守区域,与山羊草、大麦等植物的查尔酮合成酶基因相似度高;属于疏水性蛋白,Ⅲ型聚酮合酶,具有同源二聚体结构,亚细胞定位结果显示基因编码的蛋白定位于细胞质。通过对PpCHS1进化和表达分析表明,其功能可能与花发育相关并能促进黄酮类物质的合成,受紫外线和高盐的诱导,同时在外施MeJA下持续高表达,推断PpCHS1可能参与非生物胁迫与生物胁迫的防御。

【Abstract】 Based on the data of transcriptome of Kentucky bluegrass,cDNA sequence of PpCHS1 gene was cloned.The gene contains a family conserved region of CHS,and had a similarity of more than 85.4%with other plant chalcone synthase genes.The protein belonged to hydrophobic acidic protein,polyketide synthase type Ⅲ,had homologous two-polymer structure.Subcellular localization results showed that the protein encoded by this gene was localized in cytoplasm.The study of PpCHS1 expression and evolution analysis showed that the gene function was related to flower development and can promote the synthesis of flavonoids,induced by ultraviolet and high salt,in the same in the application of MeJA the expression was in high level,thus PpCHS1 may be involved in stress and biological defense to abiotic stress.
