TRAIL协同五味子乙素对脑肿瘤干细胞生长的影响
Effect of Tumor necrosis factor-related apoptosis-inducing ligand(TRAIL) combined with Schisandrin B(Sch B) on the growth of brain tumor stem cells
【摘要】 目的探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)与五味子乙素(SchB)对脑肿瘤干细胞(BTSCs)生长的影响。方法原代培养人脑肿瘤细胞,采用CD133免疫磁珠法获得BTSCs,噻唑蓝(MTT)法检测TRAIL与SchB单独或联合作用后BTSCs增殖能力的变化。结果不同浓度TRAIL作用于BTSCs均可抑制其增殖,与对照组比较,30μg/ml和100μg/ml组差异有统计学意义(P <0. 01)。不同浓度SchB作用于BTSCs时,亦均可以抑制其增殖,与对照组比较,25μg/ml、50μg/ml和100μg/ml组差异有统计学意义(P <0. 01)。不同浓度TRAIL联合100μg/ml SchB可以明显地抑制BTSCs的增殖,与对照组比较,TRAIL浓度30μg/ml和100μg/ml组差异有统计学意义(P <0. 01)。结论 TRAIL与SchB均可以抑制BTSCs的增殖,且两者可发挥明显的协同作用。
【Abstract】 Objective To investigate the effects of tumor necrosis factor-related apoptosis-inducing ligand( TRAIL)and Schisandrin B( SchB) on the growth of brain tumor stem cells( BTSCs). Methods BTSCs was obtained from primary culture of human brain tumor cells through CD133 immunomagnetic beads screening method. Thiazole blue( MTT) assay was used to detect the proliferation of BTSCs after treatment with TRAIL and SchB alone or in combination. Results Different concentrations of TRAIL could inhibit the proliferation of BTSCs. The inhibitory effects of 30 μg/ml and 100 μg/ml groups were statistically significant compared with the control group( P < 0. 01). When different concentrations of SchB were applied to BTSCs,the cell proliferation was also inhibited. The inhibitory effects of 25 μg/ml,50 μg/ml and 100 μg/ml groups were also statistically significant in comparation with the control group( P < 0. 01). Different concentrations of TRAIL( 30 μg/ml and 100 μg/ml) combined with 100 μg/ml SchB significantly inhibited the proliferation of BTSCs,and the inhibitory effects were statistically significant compared with the control group( P < 0. 01). Conclusion Both TRAIL and SchB can inhibit the proliferation of BTSCs,and the two can play a significant synergistic effect.
