构建血站NAT实验室环境监控体系的初探
A Preliminary Study on the Construction of NAT Laboratory Environmental Monitoring System of Blood Station
【摘要】 目的:建立和完善血站NAT实验室环境监控体系。方法:应用环境监控器材、选取动静态结合的模式,通过空气沉降、物表擦拭、特殊采样等方法,定期对NAT实验室进行环境监测,对监控结果结合NAT混检阳性率、拆分阳性率进行统计分析和综合评价。结果:2016年以来我室对试剂准备、样品制备、扩增检测3个区域20余监控点分批进行10次环境监测,其中2016年6月和8月均出现全自动核酸提取仪出风口监测点阳性; 2016年11月和2018年6月各为样本制备和扩增检测区内门把手监测点阳性,其余6次合格率均为100%。分析环监合格率与同期NAT质量监控指标的相关性,其中与混检阳性率r1(-0. 278)<P1(0. 437)、与拆分阳性率r2(-0. 143)<P2(0. 694),两组数据均无统计学意义。结论:偶发的设备和物表污染未对NAT检测造成明显影响,NAT实验室应在有效污染防范的基础上,逐步建立和完善环境监控体系。
【Abstract】 Objective: Establish and improve the NAT laboratory environmental monitoring system. Methods: With the application of environmental monitoring equipment and the selection of dynamic and static state mode,the NAT laboratory is regularly monitored for environmental monitoring by means of air settlement,surface wiping and special sampling. The monitoring results are combined with the positive rate of NAT mixed detection and single detection for statistical analysis and comprehensive evaluation. Results: Since 2016,we have carried out 10 environmental monitoring on more than 20 monitoring points in 3 regions,including reagent preparation,sample preparation and amplification detection. In June and August 2016,all the monitoring points of the outlet of the automatic nucleic acid extraction instrument were positive. November 2016 and June 2018,the doorknob monitoring points of reagent preparation and amplification detection regions were respectively positive,the other 6 times passed 100%. The qualified rate of environmental monitoring and the NAT quality monitoring indicators index during the same period were analyzed,the correlation to the positive rate of NAT mixed detection r1(-0. 278) < P1( 0. 437),To the positive rate of single detection r2(-0. 143) < P2( 0. 694) were not statistically significant for both groups. Conclusion: Occasional equipment or surface pollution have not significantly affected NAT detection. The NAT laboratory should gradually establish and improve the environmental monitoring system on the basis of effective pollution prevention.
