父代高脂饮食对F1代雄鼠睾丸超微结构的影响

【摘要】 目的:探讨父代高脂饮食对F1代雄性小鼠睾丸超微结构的影响。方法:将20只3周龄C57BL/6雄性小鼠随机分为正常饮食组(CD)和高脂饮食组(HFD),分别给予普通饲料和高脂饲料。连续饲养12周后,分别与8周龄正常雌性小鼠合笼繁育F1代,在F1代15周龄大时摘取睾丸组织制作电镜切片观察超微结构。结果:CD组F1代雄鼠睾丸超微结构完整,HFD组F1代雄鼠睾丸曲细精管膜基底肿胀、厚薄不均,生精细胞层数减少、紊乱,细胞间连接疏松,胞内出现电子密度高的异常结构,线粒体出现空泡样化,精子顶体空泡。结论:高脂饮食能够对F1代雄鼠睾丸形态和生精细胞造成间接损伤,影响F1代睾丸的正常生殖功能。

【Abstract】 Objective:To investigate the effects of high fat diet on the ultrastructure of the testis of F1 male mice.Methods:20 C57 BL/6 mice aged 3 weeks were divided into two groups by random number table method.High fat diet group(HFD)was fed with high fat diet while normal fat diet group(CD)was fed with normal fat diet.At the end of 12 weeks,CD/HFD F0 males were then mated with 8-week-old normal weight naturally cycling females to produce F1 offspring.At 15 weeks of age,the ultrastructure of the testicular tissue was observed by electron microscopy.Results:The ultrastructure of the testis of F1 male mice was complete in group CD,comparatively,which showed thickened and winkled basement membrane of seminiferous tubules,the layers decreased,the intercellular connections were loose,abnormal structures with high electron density appeared in the cells and vacuolization appeared in mitochondria.Conclusion:High fat diet leads to indirect damage to the testis morphology and spermatogenic cells of F1 male mice,and may affect the normal reproductive function of the testis of the F1 generation.
