氨茶碱单剂量与多剂量给药在家兔体内的药动学研究
Study on pharmacokinetics of aminophylline inrabbits by a single-dose and multi-dose administration
【摘要】 目的氨茶碱单剂量与多剂量口服给药后测定在家兔体内的药代动力学参数,为临床监测氨茶碱血药浓度提供一个简便快捷的方法。方法分别建立单波长与双波长紫外分光光度法测定氨茶碱血药浓度的方法,通过对两种方法的比较,最终确定简单快捷的单波长法(275 nm)作为测定方法。结果单剂量口服给药t1/2=(7.35±0.16)h,k=(0.094±0.020)h-1,ka=(1.44±0.40)h-1,AUC0→t=(162.81±34.56)h·μg-1·ml-1。多剂量口服后的Css=(21.36±3.05)μg/ml,AUC0→τ=(169.28±43.56)h·μg-1·ml-1。结论建立的血药浓度测定方法简便准确,相关药动学参数可为临床设计氨茶碱最佳给药剂量和给药周期提供良好的理论及实验依据。
【Abstract】 Objective To provide a simple and quick method for monitoring the blood concentration of amino-phylline inclinical practice by calculate thepharmacokinetic parameters of aminophylline through a single-dose and multi-dose administration. Methods The single and dual-wavelength ultraviolet spectrophotometry to determine aminophylline plasma concentration was established respectively. By comparing the two methods, the simple and quick single wavelength ultraviolet spectrophotometry(275 nm) was identified as the measuring method. Results After single-dose oral administration, t1/2=(7.35 ±0.16) h,k =(0.094 ±0.020) h-1, ka=(1.44 ±0.40) h-1 and AUC0 →t=(162.81 ±34.56) h·μg·ml-1. Aftermultiple-dose oral administration, =(21.36 ±3.05)μg·ml-1 and AUC0→τ=(169.28 ±43.56) h·μg·ml-1. Conclusion The method for determining the blood concentration of aminophylline was convenient and accurate. The pharmacokinetic parameters of aminophylline can provide a good theoretical and practical basis for the best drug dosage and dosing cycle.
