【摘要】 目的探讨微小RNA-502-3p(miR-502-3p)在肾癌组织中的表达及对肾癌细胞迁移、增殖的影响。方法采用荧光定量PCR检测30例肾癌及癌旁正常组织中miR-502-3p的表达,分析其与患者临床病理特征的关系。培养786-O肾癌细胞,采用miR-502-3p抑制剂(下调组)、功能类似物(过表达组)或乱序(对照组)转染细胞,Transwell实验以及划痕实验检测肾癌细胞迁移能力,CCK-8实验检测肾癌细胞增殖能力。结果肾癌组织miR-502-3p表达低于癌旁组织(P<0.01)。miR-502-3p表达水平与患者年龄、性别、病理类型和临床分期无关(P>0.05)。与对照组相比,下调组细胞迁移细胞数增加,过表达组迁移细胞数量减少(P<0.01或P<0.05)。与对照组相比,下调组肾癌细胞增殖能力增强,过表达组增殖能力降低(P<0.01)。结论 miR-502-3p在肾癌组织中低表达。miR-502-3p能抑制肾癌细胞的迁移和增殖,可能与肾癌的发生和发展有关。更多还原

【Abstract】 Objective To investigate the expression of microRNA-502-3 p(miR-502-3p)in renal carcinoma tissues and its effect on the migration and proliferation of renal carcinoma cells.Methods The expression of miR-502-3p in 30 cases of renal carcinoma and tumor-adjacent normal tissues were detected by fluorescence quantitative PCR.The relationship of miR-502-3p expression and clinicopathological characteristics was analyzed.The renal carcinoma cell 786-O was cultured and transfected by miR-502-3p inhibitor(group A),functional analogue(group B)and disorder(group C).The cell migration and proliferation were detected.Results The expression of miR-502-3p in renal carcinoma tissues was lower than that in tumor-adjacent normal tissues(P<0.01).The expression of miR-502-3p was not related to the age,gender,pathological type and clinical stages(P>0.05).Compared to group C,the number of migration cell and proliferation ability were higher in group A and lower in group B(P<0.01 or P<0.05).Conclusion The expression of miR-502-3p is downregulated in renal carcinoma tissues.miR-502-3p can inhibit the migration and proliferation of renal carcinoma cells,indicating that it may be involved in the occurrence and development of renal carcinoma.更多还原
